摘 要:目的探讨紫草素(shikonin)对体外鼻咽癌细胞增殖及凋亡的影响。方法经紫草素处理后的人鼻咽癌细胞株CNE-2和永生化非癌性人鼻咽上皮细胞株NP-69,采用CCK-8试剂盒检测细胞增殖抑制情况;在荧光显微镜下利用Hoechst33258荧光染色法观察药物处理后细胞的凋亡现象;应用流式细胞仪(FCM)分析细胞凋亡率的变化。结果不同浓度紫草素对CNE-2细胞的增殖均有抑制作用,与无药物处理对照组相比差异具有统计学意义(P<0.05或P<0.01),并呈时间和浓度依赖性,其中10μg/ml紫草素处理CNE-2细胞72 h后的抑制率高达84.64%;Hoechst33258染色检测发现典型的凋亡细胞核形态学变化;流式细胞仪检测显示,CNE-2细胞实验组的凋亡率高于对照组(P<0.05)。结论紫草素能明显抑制体外鼻咽癌CNE-2细胞的生长,呈浓度与时间依赖性,并诱导其凋亡。Objective To investigate the effects of shikonin on proliferation and apoptosis of nasopharyngeal carcinoma cell line in vitro. Methods The nasopharyngeal carcinoma cell lines CNE - 2 treated with different concentrations of shikonin as experi- ment group and the non - cancerous immortalized nasopharyngeal epithelial cell lines NP - 69 without shikonin treatment as con- trol group were involved to observe the effect of shikonin on cell proliferation using CCK - 8 assay. Cell apoptosis was observed under the fluorescence microscope with Hoeehst 33258 dye. The change of apoptosis rate was analyzed by flow cyctometry. Re- sults The growth of CNE - 2 cells was significantly inhibited by shikonin in a time - and dose - dependent manner compared with the control group( P 〈 0.05 or P 〈 0.01 ), the inhibition rate of CNE - 2 cells treated by 10 μg/ml of shikonin for 72 h was 84.64% ; After being treated with shikonin, the typical apoptotic nuclear morphological changes were observed in CNE - 2 cells under the fluorescence microscope in Hoechst 33258 staining experiment. The results of flow cytometry analysis showed that the apoptosis rate of the experiment group was higher than the control group ( P 〈 0.05 ). Conclusion It was clear that the shiko- nin can inhibit the proliferation of human nasopharyngeal carcinoma cells in a time - and dose - dependent manne and induce cell apoptosis.
