摘 要:以野生仙人掌为材料,通过单因素试验及L9(34)正交试验对仙人掌多糖的热水浸提工艺进行优化;优化工艺条件下,比较茎龄为2~4个月及茎龄大于5个月的仙人掌多糖提取结果;并通过对羟基自由基(·OH)的清除比较不同提取方法得到仙人掌多糖的体外抗氧化活性。结果表明,最佳提取工艺条件为:液料比60︰1(mL/g)、提取时间3 h、提取温度95℃,此优化条件下茎龄为2~4个月的野生仙人掌多糖提取率为30.28%±0.22%,极显著高于茎龄大于5个月的野生仙人掌多糖提取率(13.85%±0.08%);2种多糖纯度及蛋白含量无显著性差异。水提仙人掌多糖有明显的抗氧化活性,其中多糖质量浓度为7.5 mg/mL时,对·OH的清除率为57.7%,与2 mg/mL VC的·OH清除率56.0%相当。The hot waterextraction of polysaccharides^from Opuntiadillenii Haw. was optimized by using the single factor and orthogonal array design methods. Under the optimized condition, the extraction results of polysaccharides from the 2-4 months old and the more than 5 months old Opuntia dillenii Haw. were compared, and the antioxidant activities ofpolysaccharides extracted by different methods were also evaluated by the hydroxyl free radical (.OH) scavenging assay in vitro. The results showed that the optimal extraction condition was as follows: liquid-material ratio 60 : 1 (mUg), extraction time 3 h, and extraction temperature 95 %. Under the condition, the extraction rate of polysaccharides obtained from 2-4 months old Opuntia dillenii Haw. was 30.28% q- 0.22%, higher than that extracted from more than 5 months old Opuntia dillenii Haw. (13.85% ~: 0.08%). The antioxidation activity test in vitro indicated that the Opuntia dillenii Haw. polysaccharides had obvious scavenging efficiency to ~ OH. When the content of Opuntia dillenii Haw. polysaccharides reached 7.5 mg/mL, its scavenging was efficiency to "OH was 57.7% and equal to that of 2 mg/mL VC (56.0%).
