摘 要:目的:确定石荠苧总黄酮的大孔树脂吸附纯化工艺条件及其体外抑菌活性。方法:采用静态吸附和解吸实验考察不同型号大孔树脂对石荠苧总黄酮的吸附和解吸性能,确定其最佳分离纯化条件。采用二倍稀释法,检测石荠苧总黄酮对7种常见致病菌的抑菌效果。结果:HPD300大孔树脂对石荠苧总黄酮的吸附量最大,吸附率和回收率也最高。在室温下,将浓度为5.55 mg/mL(pH=3.0)的石荠苧总黄酮提取液,以2.0 mL/min速上经预处理后的HPD300大孔树脂柱,吸附后先以80 mL蒸馏水洗脱树脂柱,再用70 mL 60%乙醇(pH=8.0)以2.0 mL/min流速洗脱。此工艺条件下石荠苧总黄酮的回收率达89.12%,纯度为90.77%。纯化所得的石荠苧总黄酮对金黄色葡萄球菌、表皮葡萄球菌、痢疾杆菌具有较强的抑菌作用,其MIC分别为0.31 mg/mL、0.63 mg/mL、0.63 mg/mL。结论:HPD300大孔树脂适用于石荠苧总黄酮的分离纯化。石荠苧总黄酮对金黄色葡萄球菌、痢疾杆菌具有较强的抑菌作用。Objective:To investigate the purification and antibacterial activity of total flavonoids extracted from Mosla scabra by HPD300 macroporous resin. Method:The dynamic conditions of adsorption and separation were analyzed with HPD300 macroporous resin. The flavonoids adsorption capacity was used as the evaluation criteria. A diploid dilution method was used to evaluate the MIC of pure flavonoids. Result : HPD300 has the good performance for adsorbing and sepa- rating total flavonoids from M. scabra in the following technological condition:the concentration of the flavonoids sample extract ( pH = 3.0) was 5.55 mg/mL ; the current velocity was 2.0 mL/min ; the eluting reagent was 70 mL of 60% etha- nol ( pH = 8.0) with the speed of 2.0 mL/min. The recovery rate was 89.12% and the purity was 90.77%. The purified flavonoids had good bacteriostasic activity on Staphylococcus aureus,Shigella dysenteriae and Staphylococcus epidermidis with the MIC as 0.31 mg/mL,0.63 mg/mL,0.63 mg/mL, respectively. Conclusion:h is a simple and efficient to separate total flavonoids from M. scabra. The purified flavonoids have the obvious antibacterial activity on Staphylococcus aureus, Shigella dysenteriae and Staphylococcus epidermidis.
