中药蛇六谷提取物对人三阴性乳腺癌MDA-MB-231细胞增殖的影响

摘　　要：目的:观察蛇六谷提取物对体外生长的人三阴性乳腺癌MDA-MB-231细胞增殖的影响,寻找蛇六谷抗肿瘤的有效部位。方法:采用系统溶剂法初步提取分离蛇六谷不同有效部位,运用CCK8法和生长曲线法观察不同浓度的蛇六谷各提取物对体外培养的MDA-MB-231细胞增殖的抑制作用,并以流式细胞仪检测其对MDA-MB-231细胞周期的影响。结果:蛇六谷石油醚萃取物、高浓度蛇六谷乙酸乙酯萃取物对MDA-MB-231细胞生长有明显抑制作用,其中蛇六谷石油醚萃取物抑制作用最为明显(P<0.05);经流式细胞仪分析蛇六谷石油醚提取物则将MDA-MB-231细胞阻滞在S期,使得细胞无法完成DNA复制进入G2期。结论:蛇六谷石油醚萃取物可能是该药抗肿瘤的有效部位之一。Objective：To observe the effect of different extracts of Rhizoma Amorphophalli on the MDA - MB -231 proliferation of human triple negative breast cancer and to explore the antineoplastic parts of the plant. Methods ： Different extracts of Rhizoma Amorphophalli were extracted by the technique of solvent - refining. The effects of different dose groups of extracts on cell MDA - MB - 231 were tested by CCK8 and growth curve method. The effects on cell cycle of MDA - MB - 231 were inspected by flow cytometry. Results ： The extracts of petroleum ether and high concentration of eth- ylacetate of Rhizoma Amorphophalli had inhibitory effect on the proliferation of cell MDA- MB- 231 ,and the extract of petroleum ether had much more evident effects （ P 〈 0.05 ）. Flow cytometry detection showed the extract of petroleum ether of Rhizoma Amorphophalli can block the cell cycle of MDA - MB -231 at S period, making the cells unable to complete DNA replication into the G2 phase. Conclusion：The extract of petroleum ether of Rhizoma Amorphophalli is possibly one of antineoplastic parts of the plant.
