摘 要:目的观察Notch通路抑制剂对骨肉瘤细胞U20S的作用。方法常规培养U20S细胞,应用R04929097处理U20S细胞,通过细胞计数试剂盒(CCK-8)法观察R04929097对U20S细胞的毒性作用,通过流式细胞仪检测R04929097对U20S细胞周期的影响及诱导凋亡的作用。结果CCK-8检测结果显示不同浓度R04929097对U20S细胞有明显的抑制作用,与对照组比较差异有统计学意义(P〈0.05),流式细胞检测结果显示R04929097能促进U20S细胞凋亡而不影响细胞周期。结论Notch通路抑制剂可诱导U20S凋亡。Objective To explore the effects of γ-secretase inhibitors on U2OS cells. Methods U2OS cells cultured under normoxic conditions were treated with RO4929097. The cytotoxicity was detec ted by cell counting Kit-8 (CCK-8) kit. The change of cell cycle and apoptosis were assayed by flow c- tometry (FCM). Results The present study showed that RO4929097 has an obvious inhibition effect on U2OS cells. It had also been found that the apoptosis was aggravated while the cell cycle wasn' t disturbed by RO4929097. Conclusion The apoptosis of U2OS cells could be aggravated by γ-secretase inhibitors.
