长链非编码RNA在肝细胞肝癌组织中的表达及临床意义

摘　　要：目的探讨长链非编码RNA（LncRNA）在肝细胞肝癌（以下简称肝癌）组织中的表达及其临床意义。方法采用实时定量聚合酶链反应（Real-timePCR）法检测7种与肿瘤相关LncRNA[B细胞整合簇基因（BIC）、淋巴细胞白血病中删除基因1（DLEU1）、小核RNA看家基因5（SNHG5）、反义胰岛素样生长因子2（IGF2AS）、前列腺癌基因3（PCA3）、α-缺氧诱导因子（HIF-α）、前列腺癌基因表达标记1（PCGEM1）]在肝癌组织和癌旁组织中（n=40）的表达。结果SNHG5、IGF2AS、PCA3、HIF-α在肝癌组织和癌旁组织中的表达差异有统计学意义（P〈0．01）；其中IGF2AS为表达差异最显著的LncRNA[肝癌：10．910±1．775）比癌旁组织（13．517±3．077）]，且在肝癌组织中的表达与肿瘤数目（P〈0．05）和微血管侵犯（P〈0．01）密切相关。Cox多因素分析显示，IGF2AS存肿瘤组织中的表达是肝癌患者无瘤生存的独立预后因素。结论新的LncRNA如IGF2AS在肝癌发生发展及预后中的重要作用值得进一步研究。Objective To investigate the expression of long chain non-coding RNA （LncRNA） in hepatocellular carc.inoma tissue and its clinical significance. Methods Real-time quantitative polymerase chain reaction （ Real-time PCR） method was used to detect the expression of seven kinds of LncRNA asso ciated with tumor [ B cell integration cluster gene（BIC）, deleted in lymphocytic leukemia 1 （DLEU1）, small nucleolar RNA host genes 5 （SNHG5） , insulin-like growth factor 2,antisense （IGF2AS） , prostate cancer gene 3 （ PCA3 ） , Hypoxia inducible factor α （HIF-a） and Prostate cancer gene expression marker 1 （ PCGEM1 ） ] in liver cancer tissues and adjacent tissues （ n = 40）. Results The expression of SNHGS, IGF2AS, PCA3, and HIF-α in liver cancer tissues and adjacent tissues was statistically significantly differ ent （ respectively P 〈 O. Ol ）. IGF2AS was most significantly differentially expressed [ HCC （ 10. 910± 1. 775 ） vs. paratumor （ 13. 517 ±3. 077） ] , and its expression in liver cancer tissue was closely related to tumor numher （P 〈0. 05 ） and microvascular invasion （P 〈 O. Ol ）. Cox multi-factor analysis showed that the expression of IGF2AS in tumor tissues was an independent prognostic factors of the liver disease-free survival. Conclusion The important role of IGF2AS played in HCC development and prognosis is worthy of further study.
