大肠癌组织中肿瘤睾丸抗原前列腺相关基因4的表达及其临床意义

摘　　要：目的筛选大肠癌相关肿瘤睾丸抗原，为大肠癌肿瘤的诊断和免疫治疗提供依据。方法样本取自本院的大肠癌患者46例，包括癌组织（原发灶）、癌旁组织。提取样本核糖核酸（RNA）后逆转录合成单链互补脱氧核糖核酸（cDNA），利用聚合酶链反应（PCR）技术测定前列腺相关基因4（PAGE4）在各组织中的相对表达量并进行分析。结果有转移患者癌组织中PAGE4相对表达量（9．85×1014±3．67×10^-3）与无转移患者癌组织中相对表达量（3．93X10^-5±1．20X10^-4）差异有统计学意义（P〈0．05）；有转移患者癌组织中PAGFA表达阳性率（25％）与癌旁组织（0）比较差异有统计学意义（P〈0．05）；无转移患者癌组织中PAGE4表达阳性率（19．2％）与癌旁组织（11．5％）比较差异无统计学意义（P〉0．05）。结论PAGFA的表达可作为肿瘤转移的预测因素，转移性大肠癌者可能是针对PAGE4标靶的肿瘤免疫治疗的适宜人群。Objective To identify the cancer testis antigens associated with colorectal cancer for clinical diagnosis and immunotherapy. Methods The expression level of prostate-associated gene 4 （PAGE4） was detected by polymerase chain reaction （PCR） in the specimens, including cancer tissues, and cancer adjacent tissues from eolorectal cancer cases （ n = 46 ） in our hospital. Results There was sig nificant difference in the PAGFA expression between metastatic tissues （9. 85 x 10 ^-4 ±3.67 x 10^-3 ） and non-metastatic cancer tissues （ 3.93 x 10^ -5 ± 1.20 x 10^- 4 ） （ p 〈 0.05 ）. There was significant difference in PAGE4 positive expression rate between cancer tissues （25%） and cancer adjacent tissues （0） in meta static cancer cases （P 〈 0. 05）. There was no significant difference in PAGE4 positive expression rate be tween cancer tissues （ 19. 2% ） and cancer adjacent tissues （ 11.5% ） in non-metastatic cancer cases （P 〉 O. 05 ）. Conclusion PAGFA might be potential predictors for metastasis of colorectal cancer and cancer testis antigen-targeted immunotherapy might be indicated for those cases with metastasis.
