摘  要:目的探讨紧密连接蛋白1(Claudin-1)和血管内皮生长因子C(VEGF-C)蛋白在大肠癌发生发展中的相关性。方法应用链霉菌抗生物素蛋白-过氧化物酶(sP)法检测45例大肠癌组织、癌旁组织以及20例大肠组织中2个基因的表达,并进行相关分析。结果大肠癌组织中的C|audin-l表达阳性率明显高于癌旁组织和正常组织(60.0%、33.3%和10.0%),VEGF—C表达阳性率明显高于癌旁组织和正常组织(64.4%、26.7%和10.0%),并且两者的表达均与肿瘤分化程度、浸润深度、淋巴结转移以及临床分期密切相关,两者的表达呈直线正相关。结论Claudin-1可能通过VEGF-C信号途径参与大肠癌的发生发展。Objective To investigate the correlation of Claudin-1 and vascular endothelial growth factor C ( VEGF-C ) protein expression in colorectal cancer development. Methods The immunohisto chemical staining [ streptavid-in-peroxidase (SP) method] was done to detect the expression of Claudin-1 and VEGF-C in 45 cases of colorectal cancer tissues and the corresponding tissues adjacent to carcinoma, and 20 cases of normal colon mucosa tissues. Results The positive expression rate of Claudin-1 in cancer tissues was significantly higher than that in the tissues adjacent to carcinoma and normal tissues (60. 0%, 33.3% and 10.0% ). The positive expression rate of VEGF-C in cancer tissues was significantly higher than that in the tissues adjacent to carcinoma and normal tissues (64. 4% , 26. 7% and 10. 0% ). Both Claudin-1 and VEGF-C expression was correlated with tumor differentiation, depth of invasion, lymph node metastasis and TNM stage. The expression of Claudin-1 and VEGF-C had a linear correlation. Conclusion Claudin-1 is involved in the development of colorectal cancer possibly through VEGF-C signaling pathways.
