摘　　要：目的观察细胞窖蛋白-l（Cav-1）在人胰腺癌中的表达，探讨Cav-1存人胰腺癌中的表达及其临床意义。方法Western blot检测人胰腺癌细胞株（MDA48、MDA28等）和人胰腺导管上皮（HPDE）细胞株中Car-l蛋白的表达。免疫组织化学检测组织芯片中70例人胰腺癌和10例正常胰腺组织中Cav-1的表达，并分析其与胰腺癌临床病理特征的关系。结果Westernblot结果显示Cav-1在多种人胰腺癌细胞株中高表达。免疫组织化学检测结果显示Cav-1在人胰腺癌组织中高表达，与组织学分级、临床分期及肿瘤转移有关（P〈0．01）。结论Cav-l在胰腺癌发生、发展中具有重要作用，与胰腺癌组织学分级、临床分期及肿瘤转移密切相关。Objective This research is designed to study the expression of Caveolin-1 （ Cav-1 ） in human pancreatic cancer and to analyze its clinical significance. Methods Western blotting were used to detect protein levels of Cav-1 in human pancreatic cancer cell lines （MDA48, MDA28, et al. ） and nor- mal human pancreatic ductal epithelial （HPDE） cell line. Tissue microarray-based immunohistochemistry was used to investigate the expression of Cav-1 protein in the 70 primary pancreatic tumor and 10 normal pancreatic tissue specimens. Correlations of Cav-1 expression to histological grade, clinical stage and lymph node or distant metastasis were analyzed. Results Car-1 was markedly upregulated in most pancre atic cancer cell lines as compared with the HPDE cell line. The results of immunohistochemistry showed that the expression of Car-1 was upregulated in the pancreatic cancer tissue samples. Increased Car-1 ex pression directly correlated with poor level of tumor differentiation, late stage and lymph node metastasis or distant metastasis. Conclusion Increased expression of Cav-1 is associated with poor level of tumor differ entiation, late stage and lymph node metastasis or distant metastasis. And Cav-I may have an oncogenic function in tumorigenesis and progression in pancreatic cancer.
