摘  要:目的初步研究腺样囊性癌细胞系(SACC83)与肺高转移的腺样囊性癌细胞系(SACC-LM)中MMP-1、MMP-2、MMP-3和MMP-7的表达差异。方法通过免疫细胞化学法、RT-PCR、Western blot研究不同转移特性的腺样囊性癌细胞系SACC-LM、SACC83中基质金属蛋白酶(MMP-1、MMP-2、MMP-3、MMP-7)在mRNA和蛋白质水平的表达。结果免疫细胞化学法中MMP-1、-2、-3、-7在两种腺样囊性癌细胞均表达。在mRNA水平,MMP-1、-2、-3、-7在两种腺样囊性癌细胞的表达差异无显著性统计学意义。但是在Western blot实验中,SACCLM中MMP-1、-2、-3的蛋白质相对表达量较SACC83增多,差异有统计学意义。结论基质金属蛋白酶MMP-1、-2、-3在肺高转移特性的腺样囊性癌细胞系表达增高对于研究腺样囊性癌血行转移的分子机制具有一定的意义。Objective The purpose of this experiment was to preliminary study the expression difference of matrix metalloproteinases system (MMP-1, MMP-2, MMP-3 and MMP-7) in salivary adenoid cystic carcinoma (SACC) cell lines, the parental cell line SACC-83, with a low lung metastasis rate and its daughter cell line SACC-LM, with a high lung metastasis rate. Methods By the way of immunocytochemical method, reverse transcription-poiymerase chain reaction and Western blot we studied the expression difference of mRNA and protein level in different metastasis characteristic of salivary adenoid cyst carcinoma cell lines. R6sults Matrix metaloproteinases (MMP-1, MMP-2, MMP-3 and MMP-7) could be expressed in both of SACC83 cell lines and SACC-LM cell lines. There was no significant difference of matrix metaloproteinases expression in different metastasis characteristic of salivary adenoid cyst carcinoma cell lines in RNA level. In protein level the expression of MMP-1, MMP-2 and MMP-3 in SACCLM cell lines was significantly higher than in the SACC83 cell lines (P 〈0.05). Conclusion The higher expression of matrix metalloproteinase MMP-1, MMP-2 and MMP-3 in SACCLM maybe have significance to study metastasis of salivary adenoid cystic carcinoma.
