摘 要:目的探讨纯钛钛片表面经不同方法处理对骨髓间充质干细胞矿化的影响。方法将直径为10mm,厚度为1mm,粗糙度为0.8μm纯钛钛片分三组进行表面处理:A组TiO2砂粒喷砂HCl/H2SO4混合液酸蚀组;B组Al2O3砂粒喷砂HCl/H2SO4混合液酸蚀组;C组未处理光滑组。通过SD大鼠骨髓间充质干细胞的体外培养与鉴定,将骨髓间充质干细胞培养于三组钛片表面并进行矿化诱导,采用扫描电镜观察三组纯钛片表面形貌及各组钛片表面矿化结节的形态,体视显微镜下计数矿化结节的数量。结果三组钛片表面矿化结节的数量之间均有统计学差异(P<0.05)。结论不同处理的钛表面对于骨髓间充质干细胞具有良好的相容性;TiO2砂粒喷砂酸蚀钛表面更有利于骨髓间充质干细胞的矿化。Objective To discuss the influence of different processing to titanium surface on mineralization of marrow mesenchymal stem cells, thus to explain clearly that this method is helpful to good osseointegration and finally to early stability. Methods Pure titanium discs were prepared with 10mm in diameter, lmm thickness and 0.Sp.m roughness, divided into 3 groups.Group A: TiO2 particle blasted and HC1/H2SO4 etching group. Group B: A1203 particle blasted and HC1/H2SO4 etching group. Group C: smooth group. The detachment, cultivation, and identification of marrow mesenchyma stem cells . The morphology of marrow mesenchymal stem cells and mineralization nodules on TiO2 blasted- etched titanium discs, A1203 blasted-etched titanium discs, smooth titanium discs and coverslips surfaces observed by SEM. Stereoscope microscope was employed to count mineralized tubercle quantity in each group. Results The number of Calcium deposition on surfaces of the three groups all had statistical differences (P〈0.05). Conclusion ①TiO2 particle blasted-etched titanium surfaces, A1203 particle blasted-etched titanium surfaces and smooth titanium surfaces all had fine biological property and osteoblasts could well grow on three kinds of titanium discs surfaces; ②Pure titanium piece sand-blasted with HC1/H2SO4 dual acid etching favors the cell's mineralization, thus enhances the osseointegration on the surface.
