摘 要:目的通过体外实验研究雷公藤甲素对大鼠胰腺组织的损伤作用。方法以空白组为对照,不同浓度的雷公藤甲素作用于体外培养的胰腺组织,12 h后,MTT法检测其吸光度(A490值),计算胰腺组织存活率;碘-淀粉比色法测定胰腺组织分泌的淀粉酶含量。结果随着雷公藤甲素浓度的升高,胰腺组织的存活率逐渐降低,其IC50值为0.002 mg/ml,同时胰腺淀粉酶含量升高(与对照组比较,P<0.05)。结论一定浓度的雷公藤甲素对胰腺组织有损伤作用。Objective To study the damage of Triptolide on rat pancreatic tissue in vitro. Methods Blank group as a control, rat pancreatic tissue was treated with triptolide at different concentrations for 12 hours. The tissue survival rate was revaluated by using MTT assay, while using Iodine starch method measured amylase levels. Results The survival rate of pancreatic tissue was reduced with the triptolide concentration increasing, and the value of IC50 is 0. 002 mg/ml. Meanwhile, pancreatic tissue amylase levels were increased (compare with the control group, P 〈0. 05). Conclusion A certain concentration of triptolide might cause pancreatic damage.
