黄芩素联合阿维A酸对皮肤鳞状细胞癌细胞SCL-12凋亡及caspases的影响

摘　　要：目的:探讨黄芩素联合阿维A酸对人皮肤鳞状细胞癌细胞SCL-12的凋亡诱导作用及对caspases的影响。方法:以体外培养的SCL-12细胞为研究对象,采用酶联免疫分析法(ELISA)检测凋亡的诱导情况;吖啶橙染色证实凋亡的发生;流式细胞仪检测细胞周期;Western印迹分析caspase-8,caspase-9和caspase-3表达。结果:黄芩素、阿维A酸单独及联合应用均能不同程度的诱导SCL-12细胞凋亡及阻滞细胞周期的进程,将细胞阻滞于G1期;活化caspase-8,caspase-9和caspase-3;其中黄芩素联合阿维A酸作用最为显著,差异均有统计学意义(P值均<0.05)。结论:黄芩素、阿维A酸单独及联合应用均能诱导SCL-12细胞凋亡,两种药物的联合应用对SCL-12细胞具有协同效应,这一效应可能是通过外源性死亡受体途径和内源性线粒体途径共同介导执行。Objective：To investigate the effects of baicalein and acitretin on the apoptosis and caspases expressions in human cutaneous squamous cell carcinoma cell line SCL-12. Methods： SCL-12 cells were cultured in vitro in the presence of baiealein or aci- tretin or both. Apoptosis was assessed by enzyme-linked immunosorbent assay （ELISA） in these cells. Apoptotic cells were observed by acridine orange staining. Cell cycle analysis was examined by flow cytometry. The protein expressions of caspase-8, caspase-9 and easpase-3 in SCL-I2 ceils were measured by Western blot. Results： Baicalein or acitretin or both all could induce the cell apoptosis of SCL-12 cells, arrest them in Gl-phase, and activate caspase-8, caspase-9 and caspase-3. Baiealein combined with acitretin exerted the strongest effect on all the parameters tested （ all P 〈 O. 05 ）. Conclusion ： Baicalein or acitretin or both plays an important role in the apoptosis of cutaneous squamous cell carcinoma cells, which may be mediated through Fas-and mitochondria-way. The combination of baicalein and aeitretin has synergistic effect in the induction of apoptosis for SCL-12 cells.
