摘  要:目的:探讨鹿衔草挥发油诱导人软骨肉瘤细胞SW1353凋亡的作用机制。方法:MTT法检测鹿衔草挥发油对人软骨肉瘤细胞SW1353增殖的抑制作用;流式细胞仪检测细胞凋亡;RT-PCR检测Bcl-2,Bax基因的表达;Western Blot检测Bcl-2蛋白的表达。结果:鹿衔草挥发油在50μg/mL-150μg/mL浓度范围内能明显抑制SW1353细胞的增殖,并呈剂量和时间依赖性,IC50为106μg/mL;流式细胞仪检测的结果显示鹿衔草挥发油能诱导SW1353细胞凋亡;RT-PCR结果表明Bcl-2基因表达降低,Bax基因表达增强;Western Blot结果表明Bcl-2蛋白表达降低。结论:鹿衔草挥发油通过诱导细胞凋亡抑制SW1353细胞增殖,可能与其下调Bcl-2表达,上调Bax表达有关。Objective: To investigate the effect of pyrolae herb essential oil (PHEO) on the human chondrosarcoma cells SW1353 and study the underlying mechanism. Methods: The viability of SW1353 cells was determined by MTT assay. Flow cytometry with annexin-V/propidium was performed to determine apoptosis. The mRNA expression of Bcl-2 and Bax was measured by reverse transcription polymerase chain reaction (RT-PCR). The protein expression of Bcl-2 was detected using Western Blot. Results: After treatment with 50l.tg/mL-1501ag/mL of PHEO, PHEO reduced viability and induced apoptosis of SW1353 cells in a dose- and time- dependent manner, the 50% inhibition concentration (ICs0) was 1061ag/mL. The result showed that PHEO treatment reduced Bcl-2 mRNA expression and enhanced Bax mRNA expression in SW1353 cells, and the protein expression of Bcl-2 was reduced versus control group. Conclusion: PHEO inhibites the growth of SW1353 cells by inducing cancer cell apoptosis, which may be associated with up-regulation of Bax expression and down-regulation of Bcl-2 expression.
