摘 要:目的:探讨海带多糖(LJP)对放疗后颌下腺血管的影响。方法:70只Wistar大鼠随机分为正常对照组(N),放疗对照组(R),海带多糖高剂量(300mg/kg)+照射组(LRA),海带多糖低剂量(30mg/kg)+照射组(LRB)。照射前后3d分别给予腹腔注射0.9%氯化钠溶液和海带多糖。R组,LRA组,LRB组的大鼠头颈部予以60Coγ射线15Gy一次性照射。透射电镜观察颌下腺血管内皮细胞超微结构改变,免疫组化法检测腺体微血管密度(MVD),RT-PCR检测血管内皮生长因子(VEGF),基质金属蛋白酶9(MMP-9)及其抑制剂(TIMP-1)的表达。结果:电镜观察LRA及LRB组血管内皮细胞损伤较轻;免疫组化结果显示,与R组相比,LRA及LRB组MVD较高(P<0.05);RT-PCR检测VEGF、MMP-9在LRA及LRB组的表达高于R组(P<0.05),TIMP-1在各组表达无差异。结论:LJP可减轻射线对颌下腺血管的损伤。Objective: To explore the effects of laminaria japonica polysaccharides(LJP) on vessels of submandibular gland after radiation in rats. Methods: Seventy Wistar rats were randomly divided into normal group(N-group), radiation control group(R-group), high-dose(300mg/kg) of LJP combined radiation group(LRA-group), and low-dose(30mg/kg) of LJP combined radiation group(LRB-group). The rats were administered with normal sodium or LJP by peritoneal injection from 3 days before radiation to 3 days after radiation. All rats in R group, LRA group and LRB-group were irradiated with a single exposure of 15 Gy γ-rays induced by Cobalt 60 irradiation delivered to the head and neck area. The morphologic changes of vascular endothelial cells were observed by transmission electron microscopy. Immunohistochemical technique was used to detect micro-vessel density(MVD). Reverse transcription polymerase chain reaction was used to investigate the expression of VEGF, MMP-9, TIMP- 1. Results: From electron microscope study, the lesions of endothelial cells in LRA and LRB-group were lighter. Compared with R-group,MVD in LRA and LRB-group were higher (P〈0.05). The expression of VEGF and MMP-9 in LRA and LRB-group were higher than those in R-group(P〈0.05), and there was no significant difference among the four groups on the expression of TIMP-1. Conclusions: LJP can lighten radiation-induced injury on vessels of submandibular gland.
