摘  要:目的:研究1 8F-FB-RGD在Lewis肺癌模型体内的生物分布。方法:24只荷Lewis肺癌C57BL/6小鼠随机分为实验组和对照组(每组12只),实验组和对照组经尾静脉分别注射示踪剂18F-FB-RGD和18F-FDG,并在注药后不同时相点(30、60、90、120min)分别测量血液及各主要组织器官(心、肝、脾、肺、肾、小肠、脑、肿瘤)中的放射性浓度,获得其体内生物学分布数据。结果:(1)在1 8F-FB-RGD的生物分布研究中,肿瘤部位有相当高的放射性摄取,肿瘤对血、肌肉及肺的T/NT比值均大于2.0。(2)示踪剂1 8F-FB-RGD对包括原发肿瘤和对侧肺转移瘤都作出了清晰显像;1 8F-FDG则由于心脏高摄取及肺部的高本底而无法区分肿瘤与纵隔,特异性不强,而且未发现转移瘤,灵敏度较1 8F-FB-RGD差。结论:同1 8F-FDG相比,1 8F-FB-RGD示踪剂对肺癌显影具有更高的特异性和灵敏性,有望成为肿瘤受体显影的高敏感性特异性示踪剂,在肺癌诊断、分期、转移和疗效监测方面发挥重要作用。Objective: To investigate the biodistribution of ^18F-FB-RGD in the murine model of lung carcinoma. Methods: Twenty-four C57B/6 mice bearing the lewis lung cancer were divided randomly into experimental group (^18F-FB-RGD as tracer) and control group (^18F-FDG as tracer) (12 mice per group). The biodistribution of mice for 8F-FB-RGD was measured with well-gamma detector at 30 rain, 60 min, 90 rain, 120rain after injection from tail veins, the biodistribution of mice for ^18F-fluorodeoxyglucose (^18F-FDG) was examined at 30min, 60 min, 90min, 120rain after injection as controls. Results; 1. In the biodistribution study of ^18F-FB-RGD, considerable radioactive uptake of tumor was observed. The ratios of tumor/blood, tumor/muscle and tumor/lung were all above 2.0. 2. ^18F-FB-RGD can image clear demarcation of both the primary tumor, as well as metastases in ontralateral lung. As a comparison, fluorodeoxyglucose (FDG) scans on the same mice were not able to distinguish the primary tumor and mediastinum with lower specificity, FDG can not find the metastatic lesions masked by intense cardiac uptake and high lung background,with lower sensitivity. Conclusions: Compared to ^18F-FDG, ^18F-FB-RGD tracer has higher specificity and sensitivity to lung cancer, and plays an important role in the diagnosis, staging of lung cancer, monitoring the curative effect.
