摘 要:目的:探讨甘氨酸对重症急性胰腺炎大鼠肺损伤的保护作用机制。方法:将SD大鼠72只随机分成假手术组(A组)、重症急性胰腺炎组(B组)、甘氨酸组(C组)。重症急性胰腺炎模型通过胰胆管逆行注射5%牛磺胆酸钠诱导,甘氨酸组(C组)于造模前3、6h分别从尾静脉内注入甘氨酸(3mmol/L,0.1mL/100g)预处理,各组在造模完成后分别于6、12、24h处死动物取其肺组织,采用酶联免疫吸附法(ELISA)测定肺组织高迁移率族蛋白B-1(HMGB-1)含量,采用RT-PCR法测定肺组织髓样细胞触发受体-1(TREM-1)mRNA水平。结果:与A组比较,B组肺组织HMGB-1水平、TREM-1mRNA的表达明显增高,差异均有统计学意义P<0.05);与B组相比,C组HMGB-1水平、TREM-1mRNA的表达呈下降趋势,差异均有统计学意义(P<0.05)。结论:甘氨酸可以抑制重症急性胰腺炎肺组织中TREM-1mRNA及HMGB-1的表达,从而对重症急性胰腺炎肺损伤起保护作用。Objective: To investigate the effect of glycocine on acute lung injury induced by the severe acute pancreatitis in rats. Methods: The rats were randomly divided into three groups: sham operation rats (group A), severe acute pancreatitis group (group B) and SAP rats given the glycocine(group C). The SAP model was established by injecting 5% sodium taurocholate into biliopancreatic ducts with needle. The glycocine group was injected with glycocine through vena caudalis at 3 h, 6 h before the SAP model induced. Rats in each group were sacrificed at 6 h, 12 h, 24 h after sham operation or SAP. The concentrations of HMGB-1 were measured by ELISA, TREM-1 mRNA expressions were measured by RTPCR. Results: Compared with group A, the levels of HMGB-1 and the expressions of TREM-1 mRNA in group B were significantly elevated (P〈0.05). In group C, these items were lower than that of group B (P〈0.05). Conclusions: Glyine can decrease the expression of TREM-1 mRNA and HMGB1, then can be used for treatment of severe acute pancreatitis.
