摘 要:目的探讨参芎葡萄糖注射液对急性脊髓损伤后运动功能恢复的影响和可能机制。方法应用改良的Allen’s法复制大鼠脊髓损伤模型60只,随机分为对照组(30只)和治疗组(30只),在建模后1、7、14、21和28 d用BBB评分和斜板试验评定脊髓损伤后后肢运动功能恢复并获取损伤段脊髓标本,用免疫组织化学方法和免疫印迹法检测两组脊髓在损伤后不同时段生长相关蛋白(GAP-43)和神经丝蛋白(NF200)的表达,对BBB评分、GAP-43、NF200进行相关分析。结果 BBB评分和斜板试验结果提示,治疗组大鼠7、14、21、28 d时的神经功能恢复较对照组明显提高(P<0.05);免疫组织化学方法和免疫印迹法提示,与对照组相比治疗组在一定时间段能上调损伤脊髓区GAP-43和NF200的表达(P<0.05);在术后7 d和14 d,BBB评分与GAP-43、NF200的表达正相关,在术后7、14、21、28 d GAP-43与NF200的表达正相关。结论参芎葡萄糖注射液能促进损伤脊髓的功能恢复,其机制可能与参芎葡萄糖注射液上调损伤区脊髓GAP-43、NF200的表达有关。[ Objective ] To investigate the effects of Shenxiong glucose injection on the recovery of motor tunction and the possible mechanism after acute spinal cord injury. [Methods] 60 models of spinal cord in- jury rat were created by a modified Allen's method and randomly divided into a control group (30) and a treatment group (30). Hindlimb motor function after spinal cord injury recovery was evaluated and damaged spinal cord specimens were got through BBB score and slanting board test in the modeling after 1, 7, 14, 21, 28 d. Immunohistochemistry method and Western blot were used to examine the expression of growth-associat- ed protein (GAP-43) and neurofilament protein (NF200) in different periods after spinal cord injury, and the correlation of BBB score, GAP-43 and NF200 was also analyzed. [Results] The results of BBB score and slanting board test showed thai the recovery of neurological function in the treated group was improved obvi- ously compared with the control group after 7, 14, 21, 28 d (P〈0.05), and the results achieved by immuno- histochemistry method and Western blotting show that the expression of the area GAP-43 of spinal cord in- jury and NF200 in the treatment group could be improved compared with the control group in a certain peri-od of time (P 〈0.05), and the BBB score result is positively related with the expression after 7, 14 d, and GAP-43 expression is positively related with NF200 after 7, 14, 21, 28 d. [Conclusion] Shenxiong glucose injection can promote functional recovery after spinal cord injury, the mechanism may be related to the in- crease of GAP-43 and NF200 expression after using Shenxiong glucose injection in spinal cord damage zone.
