摘  要:目的探讨氟砷联合作用对大鼠骨骼功能的影响。方法以SD大鼠为实验动物,析因设计方法随机分成9组,每组6只,雌雄各半。用氟化钠(NaF)和亚砷酸钠(NaAsO2)灌胃染毒。分别为:对照组、低氟组(简称Flow,5mg/kg)、高氟组(简称Fhigh,20 mg/kg)、低砷组(简称Aslow,2.5 mg/kg)、高砷组(简称Ashigh,10 mg/kg)、FlowAslow组、FlowAshigh组、FhighAslow组、FhighAshigh组。酶联免疫吸附法(Elisa)测定大鼠尿Ⅰ型胶原交联氨基末端肽(UNTX)、血清骨钙素(OCN)、骨涎蛋白(BSP)浓度,双能X线骨密度仪测量大鼠双侧股骨骨密度(BMD)。结果 (1)UNTX随着氟、砷剂量增高而升高,存在剂量-效应关系,且其在FlowAslow、FhighAslow剂量组分别低于两者单独作用时的浓度之和(P<0.01)。(2)BSP、OCN随着氟剂量增高而升高,存在剂量-效应关系;BSP、OCN浓度与砷相关关系不明显(P>0.05)。(3)BMD随着氟剂量的升高而增高,但差异无统计学意义(P>0.05);BMD与砷相关关系不明显(P>0.05);与对照组相比除Aslow、FlowAshigh剂量组BMD略为降低,其余各剂量组BMD均略高于对照组,但其差异均无统计学意义(P>0.05)。结论 (1)单独氟能够促进OCN、BSP浓度的升高,氟或砷单独作用能够促进UNTX浓度的升高,在FlowAslow、FhighAslow剂量组,氟、砷对UNTX表现为拮抗作用,UNTX可作为评价氟砷暴露骨损伤联合作用的生物学标志。(2)BMD在一定程度上反映氟对骨骼的损害效应,在该暴露水平下,实验大鼠骨损伤作用类型主要表现为骨质硬化。Objective The study was conducted to explore the combinatorial effect of fluorine and arsenic on bone functions of rats. Methods SD rats were randomly divided into 9 groups, with each group consisting of 3 male rats and 3 female rats, and were ex- posed to sodium fluoride (NaF) and sodium arsenite (NaAsO2) by oral garaging. The 9 groups included a control group, a low fluorine group (or Flow 5 mg/kg), a high fluorine group (or Fhigh 20 mg/kg), a low arsenic group (or Aslow, 2.5 mg/kg), a high arsenic group (or Aslow, 10 mg/kg), a FlowAslow group, a FlowAShigh group, a FhighASlow group, and a FhighAShigh group. Enzyme-linked immunosorbent assay (ELISA) was used to determine the levels of urinary cross-linked N-telopeptide of type I collagen (UNIX), serum osteocalcin (OCN), and bone sialoprotein (BSP) in the rats, and dual-energy X-ruy absorptiometry was utilized to measure the bone mineral density (BMD) of bilateral femur of the rats. Results (1) UNTX increased with the doses of fluorine and arsenic, indicating a dose-dependent effect; and the levels of UNTX in the FlowASlow, and FhighASlow groups were lower than the combined UNTX level of the Flow and Aslow groups and Fhigh and Aslow groups, respectively (P〈0.01). (2) BSP and OCN increased with the dose of fluorine, also showing a dose-dependent effect; the correlation between the levels of BSP and OCN and arsenic were weak (P〉0.05). (3) BMD increased with the dose of fluorine, but the increase was not statistically significant (P〉0.05); the correlation between BMD and arsenic was not strong (P〉0.05); the levels of BMD in groups other than ASlow and FlowAshigh were slightly higher than that of the control group, but the differences were not statistically significant (P〉0.05). Conclusion (1) Fluorine alone can cause the increase in BSP and OCN levels, and fluorine or arsenic alone can lead to the increase in UNTX level.
