摘 要:目的探究松树皮提取物(Pine bark extract)对鱼藤酮诱导帕金森病(Parkinson's Disease,PD)小鼠神经元的保护作用及可能机制。方法成年雄性C57BL/6小鼠40只,随机分为空白对照,鱼藤酮,松树皮提取物,松树皮提取物+鱼藤酮4组。实验处理5周后,化学比色法检测脑组织中MDA、GSH含量及SOD酶活力判定氧化应激损伤水平,免疫组化分析TH、α-SYN和LC3-B阳性表达变化。结果鱼藤酮组与空白对照组及鱼藤酮+松树皮提取物组比较,小鼠中脑黑质-纹状体GSH含量和SOD活性明显减少(P<0.05),MDA含量明显增加(P<0.05);小鼠中脑黑质-纹状体TH免疫阳性神经元数目明显减少(P<0.05);鱼藤酮组与空白对照组及鱼藤酮+松树皮提取物组比较,小鼠中脑黑质-纹状体系统α-SYN和LC3-B免疫阳性表达明显增多(P<0.05)。结论松树皮提取物对帕金森模型小鼠多巴胺能神经元具有一定的保护作用,其机制可能与减轻鱼藤酮所致的氧化应激损伤有关。Objective The purpose of this study was to explore the protective effect and possible mechanism of pine bark extract on impaired neuron in rotenone-induced Parkinson's disease mouse model. Methods Forty healthy adult mice were randomly divided into four groups: control group, rotenone group, pine bark extract group, and rotenone plus pine bark extract group. After five weeks, the expression of tyrosine hydroxylase, LC3-B immune antigen, c~ -synuclein in the substantia nigra and the striatum were exam- ined using immunohistochemistry. Results Compared with the control group and rotenone plus pine bark extract group, the level of GSH (P〈0.05) and the activity of SOD (P〈O.05) were significantly decrease in rotenone group, while the level of MDA was signifi- cantly higher in substantia nigral and the striatum. Rotenone significantly decreased the number of tyrosine hydroxylase positive neurons (P〈0.05) and increased the expression of ct-synuclein and LC3-B immune antigen (P〈0.05) in substantia nigral and the striatum. Conclusion Pine bark extract can protect the substantia nigra dopaminergic neurons in rotenone-induced Parkinson's disease mouse model. The protective effect of pine bark extract may be related to the attenuation of rotenone-induced oxidative stress injury.
