摘 要:目的探讨缺血预适应(PC)对大鼠肢体缺血再灌注(LIR)后脑组织细胞色素C(CytC)的影响及其可能机理。方法雄性Wistar大鼠48只,随机分成3组:对照组(control)、模型组(LIR)和缺血预适应组(IPC),每组16只,分别复制大鼠LIR模型,测定脑组织丙二醛(MDA)和胞浆细胞色素C含量,检测线粒体Ca2+、细胞色素C含量,采用尼氏染色观察脑组织病理学改变和Western印迹法检测细胞色素C的表达变化。结果大鼠LIR后,脑组织海马区、中脑红核区神经元细胞受损,模型组胞浆CytC表达明显上调,线粒体CytC表达下降,而线粒体Ca2+含量显著增加。缺血预处理后脑组织胞浆CytC、线粒体Ca2+与模型组相比含量降低,差异显著(P<0.05),而线粒体CytC表达上升,并抑制脂质过氧化的发生。结论缺血预处理可减少LIR中脑组织线粒体释放CytC,维持线粒体Ca2+稳态,对脑组织有保护作用。[ Objective ] To evaluate the effects of ischemia preconditioning (IPC) on the expression of cytochrome C (CytC), exploring the mechanism of brain injury after hind limbs ischemia-reperfusion (LIR) in rats. [ Methods ] The Limbs isehemia-reperfusion model was established (with) in rats. 48 male rats were randomly divided into three groups: control group, LIR group and IPC group, 16 rats in each group. The content of MDA was evaluated in each groups, The histo-pathological changes were determined by Nissl staining. The content of CytC in cytoplasm was measured using ELISA kits. The contents of mitoehondrial calcium and cytochrome C were detected using ELISA kits. The expression of CytC was determined with Western-blot. [ Results ] In the LIR group, the expressions of CytC and the contents of mitochondrial calcium were significantly increased than those in the control group, following IPC the expression of CytC and the contents of mitochondrial calcium were significantly decreased than those in the LIR group(P 〈0.05). The expressions of mitochondrial cytochrome C were increased in the hippoeampus and the midbrain red nucleus region, which were the more sensitive areas to isehemie injury. [ Conclusions ] Isehemie preconditioning reduced the release of mitochondria CytC and maintained mitochondrial calcium homeostasis. It is one of the protection mechanism of IPC.
