摘 要:目的:探讨超声复合提取甘草籽黄酮及多糖的工艺优化及其抗氧化性。方法:以超声提取时间、乙醇体积分数和料液比为响应因子,黄酮及多糖提取率为响应值,采用三因素三水平的响应面分析法对甘草籽黄酮、多糖的提取工艺进行优化,初步评价甘草籽黄酮、多糖在水杨酸、邻二氮菲体系中对羟自由基的清除能力。结果:在分析各因素的显著性和交互作用后,得出甘草籽黄酮、多糖的最佳提取条件分别为:黄酮为超声提取时间29.69 min、乙醇体积分数84.59%、料液比1∶22.37(g·mL-1);多糖:超声提取时间27.9 min、乙醇体积分数86.37%、料液比1∶22.97(g·mL-1)。结论:超声复合提取甘草籽黄酮、多糖的提取工艺稳定、可行,实际1次提取率分别可达0.989%和3.628%,对羟自由基有较强的抗氧化能力。Objective:To optimize the extraction condition for flavonoid and polysaccharide complex in the licorice seeds by using ultrasonic - assisted method and determine its antioxidant activity. Method: The extracting time, eth- anol content and ratio of material to liquid were selected as response factors, and the yield of polysaccharide was used as response value. The optimum conditions for flavonoid and polysaccharide extraction were obtained based on three - factor - three - level experiment design according to the principles of response surface methodology, and their ability of scavenging hydroxyl radical with the Salicylic acid and phenanthroline system was evaluated. Re- sults : After the analysis of the significance of these influencing factors and interaction between each other, the opti- mum conditions of flavonoid were obtained as follows :extracting time 29.69 min ,alcohol concentration 84.59% ,ra- tio of material to liquid 1 : 22. 37 ( g · mL^- 1 ) and the optimum conditions of polysaccharide were : extracting time 27.9 rain, alcohol concentration 86.37 %, ratio of material to liquid 1 : 22.97 (g · mL^- 1 ). Conclusion : Ultrasonic technology of extracting flavonoid and polysaecharide of licorice seeds is stable and feasible ; the extraction rate can be up to 0. 989% and 3. 628% ,respectively. In addition,the results showed that flavonoid and polysaccharide have strong antioxidant ability for hydroxyl radical.
