乳腺癌β3-微管蛋白表达与紫杉类药物化疗耐药的相关研究

摘　　要：目的探讨乳腺癌β3-微管蛋白表达与紫杉类药物化疗耐药发生的关系。方法对42例乳腺癌患者进行2～6个周期的含紫杉类药物方案化疗，并在化疗前及化疗的第2、4、6周期末检测β3-微管蛋白的表达，分析化疗疗效与β3-微管蛋白表达的相关关系以及化疗过程中β3-微管蛋白表达的改变。结果化疗前β3-微管蛋白的平均表达率为0．456±0．320，化疗总体有效率为57．1％，tubb3表达与紫杉类药物化疗有效率呈负相关。在化疗过程中，β3-微管蛋白表达呈上升趋势，并在第4疗程达到了统计学的差异。结论β3-微管蛋白是良好的紫杉类化疗敏感性预测因子，并且可能是紫杉类继发耐药的重要原因，在化疗前及化疗过程中检测其表达情况有助于化疗方案的选择。Objective To investigate the relationship between the expression of 13 m-tubulin protein and chemoresistance to taxane in breast cancer. Methods Twenty two breast cancer patients were administrated with 2 - 6 cycles of chemo- therapy with taxoids. The changes of the expression of βⅢ-tubulin protein was detected before chemotherapy and on the end of the second, fourth, sixth cycle. The relationship of the expression of βⅢ-tubulin protein and therapeutic efficacy was analyzed. Results The average expression rate ofβⅢ-tubulin before chemotherapy was O. 456± 0. 320, overall chemotherapy effective rate was 57.1% , βⅢ-tubulin expression level was negatively correlated with the sensitivity to tax- ane. During chemotherapy, the expression levels of βⅢ-tubulin showed an upward trend. Conclusion The expression levels of βⅢ-tubulin have a good value in predicting the sensitivity to taxane, and might be an important reason for the secondary drug resistance to taxane. To detect its expression will help the selection of chemotherapy regimen before or dur- ing chemotherapy.
