色氨酸羟化酶2基因多态性与晚发性抑郁症及其疗效的关联研究

摘　　要：目的探讨色氨酸羟化酶2(TPH2)基因多态性与晚发性抑郁症(LOD)及其疗效的关系。方法采用聚合酶链式反应-限制性片段长度(PCR-RFLP)法完成191例晚发性抑郁症(研究组)和162例正常对照者(对照组)的TPH2基因rs4290270、rs7305115两个位点多态性的测定。研究组在基线及治疗后6周评定17项汉密尔顿抑郁量表(HAMD-17)以评估临床疗效。结果 (1)TPH2基因rs4290270和rs7305115两位点基因型和等位基因频率在研究组和对照组的分布差异无统计学意义(P>0.05);(2)单倍型分析显示,rs4290270-rs7305115两位点单倍型在研究组和对照组之间分布的差异无统计学意义(P>0.05);(3)治疗后第6周末达痊愈组和非痊愈组及与对照组的TPH2基因rs4290270、rs7305115多态性的基因型和等位基因频率分布差异无统计学意义(P>0.05);治疗6周有效组和无效组及与对照组的rs4290270、rs7305115两位点基因型和等位基因频率分布差异无统计学意义(P>0.05)。结论 TPH2基因rs4290270,rs7305115多态性可能与晚发性抑郁症及抗抑郁剂疗效无明显关联。Objective To investigate the association of polymorphisms in tryptophanhydroxylase enzyme 2 （TPH2） gene with late-onset depression （LOD） and efficacy of antidepressants in Chinese Han population. Methods Polymorphisms of rs4290270 and rs7305115 in TPH2 gene was genotyped by using Polymerase Chain Reaction-Restriction Fragment Length Polymorphism （PCR-RFLP） in 191 LOP patients （study group） and 162 healthy controls （control group）. Patients in study group were assessed with Hamilton Depression Scale-17 items （ HAMD-17 ） at baseline and at the end of the 6th week of treatment to evaluate the efficacy of antidepressants. Results （ 1 ） There were no significant differences in genotypic frequency and allelic frequency of rs4290270 and rs7305115 in TPH2 gene between study group and control group （P 〉 0.05 ）. （2） No significant difference was found in frequency of rs4290270-rs7305115 haplotype between the two groups （P 〉 0.05）. （3） There were no significant differences in distribution of genotypes and alleles of rs4290270 and rs7305115 among remissioners, non-remissioners and healthy controls, as well as among responders, non-responders and healthy controls （P 〉 O. 05 ）. Conclusion The polymorphisms of rs4290270 and rs7305115 in TPH2 gene are unlikely associated with the pathogenesis of LOD and the efficacy of antidepressants.
