摘  要:目的探讨原花青素(PC)对2型糖尿病大鼠局灶性脑缺血再灌注后脑组织Bcl-2及Caspase-3蛋白表达的影响。方法选用健康雄性Sprague-Dawley大鼠,体质量200~250 g,常规饲料适应性饲养1周,然后喂饲高脂高糖饲料4周,诱导高胰岛素血症和胰岛素抵抗后,按25 mg/kg体质量的剂量一次性尾静脉注射链脲佐菌素(STZ)溶液建立2型糖尿病大鼠模型,造模成功后将大鼠随机分为假手术组、脑缺血再灌注组和PC预处理组,每组6只大鼠,采用线栓法建立局灶性脑缺血再灌注模型。PC预处理组于大鼠术前1周将PC(100mg/kg)粉剂用蒸馏水配制成悬浮液灌胃,每天1次,并于术前1 h再灌胃1次。应用免疫组化法观察缺血60min再灌注24 h大鼠额顶部皮质Bcl-2和Caspase-3蛋白表达。结果与假手术组比较,脑缺血再灌注组脑组织的Bcl-2和Caspase-3表达增加(均P〈0.05);与脑缺血再灌注组比较,PC预处理组Bcl-2表达显著增加(P〈0.05),Caspase-3蛋白表达明显减少(P〈0.05)。结论 PC具有增加2型糖尿病大鼠局灶性脑缺血再灌注后脑组织Bcl-2蛋白表达的作用,同时也具有降低脑组织Caspase-3蛋白表达的作用。[ Objective ] To investigate the procyanidins (Procyanidins, PC) Bcl-2 and Caspase-3 protein expres- sion in brain tissue of type 2 diabetic rats after focal cerebral ischemia and reperfusion. [Methods] A healthy male Sprague-Dawley rats, body weight (200-250) g, and the adaptability of conventional feed fed for one week and then fed high-fat high-sugar diet for four weeks after the induction of hyperinsulinemia and insulin resistance, according to dose of 25 mg/kg body weight of a one-time tail vein injection of streptozotoein (STZ) solution to establish the type 2 diabetic rat model, the model was successful later, the rats were randomly divided into sham group, cerebral ischemia and reperfusion group and PC pretreatment groups of six rats, the suture method to establish a focal cere- bral isehemia and reperfusion model. In the first week, PC pretreatment group rat preoperative PC (100 mg/kg) pow- der preparation with distilled water suspension orally, once a day, orally 1 h before surgery 1. Immunohistochemical staining was observed ischemia 60 min reperfusion for 24 h in rat frontal and parietal cortex of the Bcl-2 and Cas- pase-3 protein expression. [ Results ] Compared with the sham group, cerebral ischemia and reperfusion of brain tissue Bcl-2 and Caspase-3 expression was increased (P 〈0.05); cerebral ischemia and reperfusion group, PC pre- treatment group, Bcl-2 expression a significant increase (P 〈0.05), caspase-3 protein expression was significantly reduced (P 〈0.05). [ Conclusions ] PC with type 2 diabetic rats with focal cerebral ischemia-reperfusion brain tissue of the role of Bcl-2 protein expression, while also reducing Caspase-3 protein expression in the role of the brain tissue.
