摘  要:目的研究丙戊酸钠对Burkitt淋巴瘤Raji细胞增殖的影响及其机制。方法细胞计数法检测不同浓度丙戊酸钠处理Raji细胞后细胞的增殖活性,制作生长抑制曲线。流式细胞仪检测细胞周期和凋亡率。Western blot检测细胞周期蛋白cyclin D1和p21、凋亡抑制基因survivin的变化。结果丙戊酸钠能明显抑制Raji细胞的增殖,其作用表现为剂量依赖性和时间依赖性(P<0.01)。流式细胞仪检测发现以3 mmol/L的丙戊酸钠处理Raji细胞后细胞阻滞于G0/G1期,其凋亡率呈时间依赖性上升(P<0.01)。Western blot检测发现3 mmol/L丙戊酸钠处理后Raji细胞cyclin D1表达明显下调而p21表达明显上调(均P<0.05)。survivin表达显著下降(P<0.01)。结论丙戊酸钠对人Burkitt淋巴瘤Raji细胞具有明显的增殖抑制作用,其作用机制与细胞周期和诱导凋亡有关。Objective To investigate the effects of sodium valproate (VPA) on the proliferation of Burkitt's lymphoma Raji cells and its mechanism. Methods Raji cells were treated with different concentrations of VPA in vitro. Cell proliferation was measured by cell counting. Cell cycle and apoptosis was determined by flow cytometry. The expression of cyelin D1, p21 and survivin was detected by Western blot. Results VPA inhibited the growth of Raji cells in a dose- and time-dependent manner(P 〈 0.01 ). Raji cells were arrested in G0/G1 stage and apoptosis rate increased time- dependently(P 〈0.01 ). The expression of eyclin Dl and survivin was decreased and p21 expression was increased 'after VPA treatment in a time-dependent manner ( all P 〈 0.05 ). Conclusion VPA can inhibit the proliferation of Burkitt' s lymphoma Raji cells in vitro ,which is associated with cell cycle and apoptosis.
