胃泌素与骨桥蛋白在胃癌及癌前病变组织中表达的相关性

摘　　要：目的:探讨胃泌素(gastin,Gas)和骨桥蛋白(osteopontin,OPN)在胃癌组织中表达的相互关系.方法:取正常胃黏膜(normal gastric mucosa,NGM)、不典型增生(dimethyl sulfoxide,Dys)及胃癌组织(gastric cancer,GC)各20例,采用SP免疫组织化学法检测各组织中Gas、OPN蛋白的表达变化.结果:免疫组织化学结果显示:Gas的表达在胃癌组和不典型增生组均高于正常组,差异存在统计学意义(P<0.05),胃癌组高于不典型增生组(P<0.05);OPN的表达在胃癌组和不典型增生组均高于正常组(P<0.05),胃癌组高于不典型增生组(P<0.05).经Spearman等级相关分析结果显示:胃泌素与OPN表达呈正相关(r=0.483,P<0.05).OPN表达与胃癌分期、浸润深度及有无远处转移有关(P<0.05),随胃癌的进展和浸润深度表达增多(P<0.05),与分化程度、性别、年龄无关.结论:OPN与Gas在胃癌组织中均表达增加,OPN表达与Gas表达呈正相关关系.胃泌素诱导OPN表达增强,可能是胃泌素促进胃癌发生发展的机制之一.AIM： To explore the correlation between the expression of gastin and osteopontin （OPN） in gastric cancer. METHODS： The expression of gastin and OPN was determined by immunohistochemistry in 60 gastric specimens including 20 cases of nor- mal gastric mucosa, 20 cases of dysplasia and 20 cases of gastric carcinoma. RESULTS： The expression levels of OPN and gastin had an increasing tendency from normal gastric rnucosa to precancerous lesions and gas- tric carcinoma （P 〈 0.05）. Correlation analysis showed that there was a positive correlation between the expression of gastin and OPN. The expression of OPN was significantly related totumor differentiation and stage （P 〈 0.05 for both）, but not to age or sex. CONCLUSION： OPN and gastrin expression is increased in gastric cancer. There was a positive relationship between the expression of gastin and OPN. Up-regulated expression of OPN may be the reason why gastin plays a key role in the development of gastric cancer.
