摘 要:目的对观辣树叶进行急性毒性及遗传毒性研究,保障观辣树叶的食用安全,为观辣树叶直接作为食品提供安全性评估资料。方法采用急性毒性试验、Ames试验、小鼠骨髓细胞微核试验和小鼠精子畸形试验。小鼠急性毒性试验剂量10.0g/kg体重,观察14d内动物的死亡情况;Ames试验分5个剂量,分别为每皿8、40、200、1 000和5 000μg,接种后,在37℃下培养48 h,计数每皿的回变菌落数;小鼠骨髓细胞微核试验和小鼠精子畸形试验均设3个剂量组,分别为0.83、1.67和3.33g/kg体重,分别观察骨髓涂片中细胞的微核发生率和精子涂片中精子畸形类型及相应的数量。结果观辣树叶雌雄性小鼠经口最大耐受剂量(MTD)>10.0g/kg体重;Ames试验、小鼠骨髓细胞微核试验及小鼠精子畸形试验三项遗传学毒性试验结果都为阴性。结论观辣树叶急性毒性分级为实际无毒级,在本研究的剂量范围内未见明显遗传毒性作用。Objective To evaluate the acute toxicity and genotoxicity of moringa oleifera lam leaves. Ensure safety Moringa oleifera lam leaves, for the Moringa oleifera lam leaves directly as food safety evaluation data provided. Methods The acute toxicity test,Ames test,micronucieus test and sperm shape abnornality test were performed to evaluate the acute toxicity and genotoxicity of Moringa oleifera lam leaves in Hainan. The dose of10.0g/kg bw was chosen for the acute toxicity tests,and the mice were observed for 14 days. The doses of 8,40,200,1 000 and 5 000μg /plate were chosen for the Ames test. The treated plates were cultured at 37 ℃ for 48h,the number of bacterial colony was counted. The doses of 0.83,1.67and 3.33g/ kg were used for the micronucleus test. The number of micronucleus of bone marrow cell was counted,the types and numbers of sperm shape abnormality were assessed. Results The oral MTD of Moringa oleifera lam leaves on male or female mice was over 10.0g/kg.bw in this experiment condition. Ames test,micronecleus test and sperm shape abnormality test were negative. Conclusion Moringa oleifera lam leaves in Hainan was considered non-toxic chemical according to acute toxicity and showed no genotoxicity in this experimental design.
