Cu、Pb联合毒性对泥鳅卵细胞基因表达影响的初步研究

摘  要:目的以不同质量浓度梯度Cu2++Pb2+(5.00+0.25)mg/L、(10+0.50)mg/L、(15+0.75)mg/L联合染毒,研究重金属离子对泥鳅卵细胞Bax和Bcl-2蛋白表达的影响。方法采用静水生物测试以及免疫组化法。结果各不同剂量Cu2++Pb2+组均可使泥鳅卵细胞Bcl-2蛋白的表达量增加,具有显著性差异(P<0.05),其中以Cu2++Pb2+中等剂量组的表达量为最高;而Bax蛋白在卵细胞的表达量则下降,且各含量组无显著差异(P>0.05)。结论重金属Cu2++Pb2+联合染毒,可诱导泥鳅卵细胞凋亡,提高Bax和Bcl-2蛋白的表达量,并且可能参与泥鳅卵细胞凋亡的调控。Objective The impacts of the expression of Bax and Bcl -2 proteins on loach oocytes were studied by means of joint attack of combined toxicity with different doses of Cu2. plus Pb2+ (5.00 ± 0. 25 ) mg/L, ( 10 ±0. 50) mg/L and ( 15±0. 75 ) mg/L, respectively. Methods by means of the indoor -static- aquatic test and immunohistochemical method. Results The results showed that the different doses of Cu^2+Pb^2+ can make the expression of Bcl - 2 increased quantitatively in all groups, with the significant difference ( P 〈 0. 05 ), of which the expression in the group of medium dose ( 10 ± 0.50 ) mg/L reached a highest volume. While the expression volume of Bax in the oocytes declined, and the concentration in each group were not of significant difference (P 〉 0. 05). Conclusion The joint toxicity of heavy metal Cu^2+Pb^2+ can induce the oocytes apoptosis, and increase the expression of Bax and Bcl -2, and they also may participate in the regulation of the oocytes apoptosis.
