散瘀和胃汤对大鼠胃癌前病变的影响研究

摘　　要：目的探讨散瘀和胃汤对大鼠胃癌前病变let-7a相对表达量和胃癌前病变发生率的影响。方法采用随机数表法将60只Wistar大鼠分为对照组(11只)和造模组(49只),从造模组随机抽取5只大鼠做病理学检测,以确定造模成功。造模成功后将造模组大鼠随机分为模型组、高剂量组(散瘀和胃汤11.50 g·kg-1·d-1)、中剂量组(5.55 g·kg-1·d-1)、低剂量组(2.75 g·kg-1·d-1),每组11只。应用RT-PCR技术检测各组let-7a相对表达量,并在光镜下观察各组胃黏膜病理组织学改变及胃癌前病变发生率。结果模型组let-7a相对表达量较对照组低(t=-7.857,P=0.000),中、高剂量组let-7a相对表达量(t=3.854、7.959,P=0.001、0.000)较模型组高。中、高剂量组胃癌前病变发生率(χ2=7.90、10.29,P=0.037、0.011)较模型组低。结论 let-7a在胃癌前病变组织中呈低表达。中、高剂量散瘀和胃汤能够提高let-7a的表达量,减少胃癌前病变发生率,散瘀和胃汤可能通过增高胃黏膜组织let-7a的表达量而防止癌变。Objective To explore the effect of Sanyu hewei soup on expression of let-7a in rats with gastric precancerous lesions.Methods A total of 60 Wistar rats were selected as study subjects,then the Wistar rats were divided into 2 groups randomly：control group（11 rats） and model group（49 rats）.In order to confirmed whether the model was built successfully,5 rats of model group were selected randomly to be done pathology detection.After the model was built successfully,the rats of model group were divided into 4 groups randomly：model of let-7a in model control group was significantly lower than that in the control group（t=-7.857,P=0.000）.The expression of let-7a was significantly higher in middle dose and high dose treatment groups than in model control group（t=3.854,P=0.001;t=7.959,P=0.000）,and the incidence rates of precancerous lesions of gastric canner in middle dose and high dose treatment groups were significantly lower than that in model control group（χ2=7.90,P=0.037;χ2=10.29,P=0.011）.Conclusion The expression of let-7a was significantly lower in tissue with precancerous lesions of gastric canner.The high and middle dose of Sanyu hewei soup could up-regulate the expression of let-7a,and reduce the incidence of precancerous lesions of gastric canner significantly.Sanyu hewei soup may inhibit gastric carcinogenesis and its progression through increasing the expression of let-7a.
