胃癌、癌旁病变中p53与Bcl-2的作用及其相互关系

摘　　要：目的 :探讨 p5 3和Bcl- 2基因在胃癌和癌旁病变中的作用及其相互关系。方法 :应用SP免疫组化法 ,检测 33例胃癌、17例癌旁病变和 13例正常胃粘膜中p5 3和Bcl- 2蛋白表达。结果 :p5 3蛋白在正常胃粘膜未见表达 ,胃癌 p5 3阳性表达率和阳性细胞密度均高于癌旁病变 ,两组间存在显著性差异 (P <0 .0 5 )。正常胃粘膜中Bcl- 2仅 1例表达 ,癌和癌旁病变中Bcl- 2蛋白表达分别高于正常胃粘膜 (P <0 .0 5～ 0 .0 1) ,胃癌与癌旁病变间Bcl- 2表达无显著性差异 (P >0 .0 5 )。p5 3和Bcl- 2蛋白表达与胃癌的分化程度、病理分期和淋巴转移无明显相关性。虽 p5 3阳性组Bcl- 2表达高于 p5 3阴性组 ,但无统计学意义。结论 :p5 3和Bcl- 2蛋白在癌旁病变中均有表达 ,表明两基因异常是胃癌发生的早期分子生物学事件。p5 3与Bcl- 2基因在胃癌和癌旁病变中有协同作用 ,但未见二者之间的负性调控作用。Objective:To explore the function and their interrelationship of P53 and Bcl-2 gene in gastric carcinoma and adjacent tissue.Methods:S-P immunohistochemistry was used to examine the expression of P53 and Bcl-2 in 33 gastric carcinoma,17 adjacent tissue and 13 normal mucosa.Results:The expression of P53 protein in normal stomach mucosa are not seen.The positive expression rate and positive cell density of P53 in gastric carcinoma are higher than that of pathological changes adjacent the carcinoma ( P <0.05).The expression of Bcl-2 protein in both gastric carcinoma and adjacent tissue are separately higher than that in normal gastric mucosa( P <0.05～0.01).There is no statistical significance for the expression of Bcl-2 in gastric carcinoma and adjacent tissue ( P >0.05).The protein expression of P53 and Bcl-2 has no relativity to the tumor differentiation degree,lymph node metastasis and pathological stage( P >0.05).Though the postive expression of Bcl-2 in P53 positive group are higher than that of P53 negative group,but there is no statistical significance between them.Conclusions:The expression of both P53 and Bacl-2 protein in adjacent tissue shows that abnormal of P53 and Bcl-2 genes are an early molecular biological incident of gastric carcinoma.P53 and Bcl-2 genes have synergistic effect in gastric carcinoma and adjacent tissue,but there are not the negative adjusting function between of P53 and Bcl-2.
