摘 要:目的:测定当归配方颗粒中阿魏酸含量、水分和浸出物含量,为科学评价及有效控制其质量提供可靠方法。同时,进行当归配方颗粒及当归饮片的质量比较。方法:提取方法、色谱方法均在2010版(《中国药典》基础上进行了优化。高效液相色谱法,色谱柱:KromasilC18色谱柱(4.6mm×250mm,5μm),流速:1.0mL/min,检测波长:316nm,柱温:28℃,流动相:乙腈-0.05%磷酸水梯度洗脱系统。结果:阿魏酸在0.033-0.165μg忙0.9998,n=5)之间有良好的线性关系,平均加样回收率为98.78%,RSD=0.99%。结论:当归配方颗粒在各项含量指标上都符合当归的质量要求,阿魏酸含量高于当归饮片。采用HPLC方法测定当归配方颗粒中阿魏酸的含量,操作简单,可靠,重现性好,可作为当归配方颗粒质量控制的有效方法。Objective: To provide reliable methods for scientific evaluation and effective control of the quality of Dangui (angelic sinensis) granules by determining the contents of ferulic acid, moisture and the contents of ex- tracts in the granules. Meanwhile, to compare the quality of Dangui granules and its prepared slices. Methods: The extracting method and chromatographic process were different from the methods in pharmacopeia of 2010 edition, which were the optimization of the methods in pharmacopeia. HPLC was adopted, chromatographic column: Kro- masil C18 (4.6 min×250 mm, 5 μm), flow rate: 1.0 mL/min, detection wavelength:316 nm, the temperature:28 ℃, mo- bile phase: acetonitrile-0.05% phosphoric acid in water gradient elution system. Results: Ferulic acid showed better linear relationship in the range between 0.033 and 0.165 μg (r=0.999 8, n=5). Average recovery rate was 98.78%, RSD=0.99%. Conclusion: Dangui granules could meet quality requirement of Dangui in the contents of different in- dexes, the contents of ferulic acid are higher than these in prepared slices of Dangui. Meanwhile, according to the re- sults, it is simple, reliable and reproducible to determine the contents of ferulic acid in Dangui granules by HPLC, which is an effective method for quality control of Dangui granules.
