摘  要:目的探讨在豚鼠耳蜗缺血再灌注损伤(ischemia-reperfusion injury,I/RI)后盐酸椒苯酮胺(piperphentonamine hydrochloride,PPTA)对蜗内含半胱氨酸的天冬氨酸蛋白水解酶-1(cysteinyl aspartate specific proteinase-1,Caspase-1)的影响。方法将24只豚鼠随机分为4组,每组6只,分别为正常组、假手术组、I/RI组、PPTA组。PPTA组在豚鼠I/RI后经股静脉注射PPTA,I/RI组以生理盐水代替PPTA注射。RT-PCR检测蜗内Caspase-1的mRNA表达,组间比较检测结果。结果各组耳蜗Caspase-1的mRNA表达量比较:I/RI组显著高于正常组、假手术组和PPTA组(P<0.001);PPTA组显著高于正常组和假手术组(P<0.001);正常组和假手术组间差异无统计学意义(P>0.05)。结论 PPTA可能通过抑制Caspase-1 mRNA的表达,从而减少细胞凋亡来对I/RI耳蜗起保护作用。Objective To analyze the effect ofpiperphentonamine hydrochloride (PPTA) on cysteinyl aspartate specific proteinase-1 (Caspase-1) in cochlear of guinea pigs with ischemia reperfusion injury (FRI). Methods 24 guinea pigs were randomly divided into normal group, sham operation group, IR/I group and PPTA group, with 6 guinea pigs in each group. We injected PPTA into vena femoralis of guinea pigs in PPTA group, and injected normal saline in I/RI group. The mRNA of Caspase-1 in cochlear was detected by RT-PCR, and the data were compared among 4 groups. Results The expression ofCaspase-1 mRNA of cochlear in IR/I group was significantly higher than that in normal group, sham operation group and PPTA group (P 〈0.001), and expression of Caspase-1 mRNA of cochlear in PPTA group was significantly higher than that in normal group and sham operation group (P 〈0.001), but there was no significant difference between the normal group and the sham operation group (P 〉0.05). Conclusions PPTA may prevent the cochlear from IR/I through inhibiting the expression of Caspase-1 and then decreasing the apoptosis.
