摘  要:目的探讨抑制Smad7基因表达对PC12细胞氧糖剥夺(OGD)损伤的影响及其机制。方法利用PC12细胞OGD模型体外模拟神经细胞缺血性损伤,通过RNA干扰技术,沉默Smad7基因表达,实时定量PCR、Western blot检测Smad7基因的表达情况,MTT检测下调Smad7基因表达对细胞OGD损伤存活率的影响,Western blot检测Activin A(ActA)蛋白的表达情况。结果体外合成的siRNA转染可下调Smad7基因转录及蛋白水平的表达;经OGD16h,阳性转染组细胞存活率较阴性组显著增加,阳性转染组细胞ActA蛋白表达较阴性组显著升高。结论Smad7可负向调控缺血损伤诱导的ActA/Smads通路激活过程,抑制其表达能够发挥神经保护作用。Objective To explore the effect of silencing Smad7 gene in PC12 cells treated by the oxygen-glucose dep-rivation (OGD)and the related mechanism.Methods PC12 cells exposed to OGD was used as the model of neurons subjected to ischemic injury in vitro.SiRNA was applied to reduce the expression of Smad7 gene,which was detected by Real time RT-PCR and Western blot.The survival rate of cells after OGD was detected by MTT.Western blot was used to test the expression of Activin A.Results The expression of Smad7 was significantly reduced after siRNA transfec-tion.The survival rate of cells in OGD16h combined with Smad7 gene silencing group was higher than that in OGD16h group.The expression of ActA protein was increased in Smad7 gene silencing group after OGD16h treatment compared to single OGD treatment.Conclusion Smad7 gene silencing can increase the survival rate of PC12 cell to OGD injury and enhance the activation of ActA/Smads.
