Anti-adhesion and anti-invasion effects of 3,4′,5-trihydroxystibene-3-β-mono-D-glucoside on human colorectal carcinoma cell lines in vitro

摘  要:目的 :3,4′ ,5 三羟基芪 3 β 单 D 葡萄糖苷 (3,4′ ,5 trihydroxystibene 3 β mono D glucoside ,THMG)抗粘附抗浸润作用及其对大肠癌细胞Ⅳ型胶原酶和整合蛋白α6 β4(laminin)受体的影响。 方法 :用流式细胞术等细胞生物学技术检测 4种大肠癌细胞 (HR 8348,Hce 86 93,HT2 9,LoVo)在 0 4、0 8、1 6、3 2mmol L等浓度THMG作用下的粘附率、浸润力、Ⅳ型胶原酶及laminin受体表达。结果 :在 3 2mmol L浓度时THMG对HR 8348、Hce 86 93、HT 2 9、LoVo粘附抑制率分别为 75 6 8% ,6 9 43% ,6 7 5 5 % ,78 78% ,浸润抑制率分别为 82 6 7% ,85 71% ,89 6 5 % ,92 6 8% ,且随着浓度增大 ,粘附和浸润抑制率提高。 3 2mmol LTHMG对HR 8348、Hce 86 93、HT 2 9、LoVo的laminin受体表达抑制率分别是 35 12 % ,36 84% ,41 5 4% ,49 5 7% ,对Ⅳ型胶原酶表达抑制率分别为 2 8 75 % ,30 6 8% ,35 6 3% ,2 2 0 3%。粘附抑制与laminin受体表达抑制呈显著性相关 (r=0 814,P <0 0 1) ,浸润抑制与Ⅳ型胶原酶表达抑制呈显著性相关 (r=0 743,P <0 0 5 )。结论 :THMG可能通过抑制癌细胞laminin受体表达而发挥抗粘附效应 。Purpose:To investigate anti adhesion and anti invasion effects of 3,4′,5 trihydroxystibene 3 β mono D glucoside (THMG) and expression of laminin receptor and type Ⅳ collagenase on the colorectal carcinoma cell lines in vitro. Methods:Four cell lines (HR 8348,Hce 8693,HT29 and LoVo) from human colorectal carcinoma were treated with 0.4,0.8, 1.6 and 3.2 mmol/L of THMG. Adhesion and invasion were assayed by the modified human amniotic basement membrane model and the Boyden chamber model. Laminin receptor and tyep Ⅳ collagenase were detected by flow cytometry.Results:At the experimental concentrations THMG had significant effects in terms of adhesion and invasion on four colorectal carcinoma cell lines (HR 8348, Hce 8693, HT29 and LoVo), with adhesion inhibition rate of 75.68%,69.43%,67.55%,78.78%,at the concentration of 3.2 mmol/L,and with invasion inhibition rate in matrigel invasion assay of 82.67%, 85.71%, 89.65%, 92.68%, for those four cell lines. The adhesion inhibition rate and invasion inhibition rate were raised according to the concentrations of THMG. Laminin receptor inhibition rates by THMG at the concentration of 3.2 mmol/L were 35.12%, 36.84%, 41.54%,49.57%,and type Ⅳ collagenase inhibition rate 28.75%, 30.68%, 35.63%, and 22.03%,respectively. Conclusions:THMG, an effective component from Chinese herb ( Polygonum cuspidatum ), have anti adhesion and anti invasion effects by inhibiting expression of laminin receptor and type Ⅳ collagenase on the colorectal carcinoma cell lines.
