神经病理性疼痛脊髓背角GLT-1及GLAST表达的变化  被引量:2

摘  要:目的:观察神经病理性疼痛大鼠脊髓背角GLT-1及GLAST表达变化对疼痛的影响.方法:选取雄性SD大鼠80只随机分为4组,A组(n=10)为L5背根切断假手术组,B组(n=30)为L5背根切断组,C组(n=10)为坐骨神经周围植管与药物溶剂组,D组(n=30)为坐骨神经周围植管与给药组.通过机械刺激的行为学观察确立病理性神经痛形成过程,采用免疫组化进行细胞定量.结果:A及C组胶质细胞GLT-1及GLAST表达无变化.B、D组痛敏形成后脊髓背角胶质细胞GLT-1及GLAST的胞内外表达未发生改变;B及D组小胶质细胞GLAST术后4d表达增加至7d后逐渐下降,而星型胶质细胞GLT-1术后6d表达明显增加,直至14d仍持续增高.结论:脊髓背角胶质细胞GLT-1及GLAST不存在胞吞现象,小胶质细胞参与启动病理性神经痛但不维持其发展,星型胶质细胞主要维持病理性神经痛发展,TNF-α介导病理性神经痛大鼠脊髓背角GLT-1及GLAST表达并诱导胶质细胞参与和维持病理性神经痛.Objective:To observe the effect of the expression of GLT-1 and GLAST in spinal cord dorsal cord with neuropathic pain.Methods:Eighty male SD rats were randomly divided into four groups.Group A (n =10) was treated with sham operation with L5 dorsal root transecfion,group B (n =30) underwent L5 dorsal root transection,group C (n =10) received intubation and medicine solvent injection around the sciatic nerve and group D received intubation around the sciatic nerve and TNF-α administration.Mechanical stimulation was adapted to establish the formation process of neuropathic pain and immunohistochemistry was applied to quantify the related substances and cells.Results:No changes appeared in the expression of GLT-1 and GLAST in groups B and D after neuropathic pain was established.The expression of GLT-1 in microglial cells increased at 6 days after operation,and continued to increase through 14 days in groups B and D.GLAST of astrocytes showed increased expression at 4 days after operation until the 7 days when the expression began to decrease gradually.There were no changes of GLT-1 and GLAST in the glial cells in groups A and C.Conclusion:There is no endocytosis phenomenon of GLT-1 and GLAST in glial cells.The microglial cells are involved in the induction ofneuropathic pain but do not sustain the pain development,and the astrocytes play a role in the sustainment of pain.TNF-α mediates the expression of GLT-1 and GLAST in spinal cord dorsal horn and induces glial cells to inifiate and sustain neuropathic pain.
