摘 要:目的探讨cortactin在结直肠癌组织、癌前病变(腺瘤性息肉)、癌旁正常组织中的表达及其意义,结直肠癌细胞cortactin表达水平与细胞分裂状态的关系。方法采用免疫组织化学方法,检测cortactin在结直肠癌组织、癌前病变(腺瘤性息肉)、癌旁正常组织中的表达。用Kaplan—Meier法比较cortactin不表达或者低表达患者组与高表达患者组5年生存率。分析cortactin在结直肠癌组织中的表达与临床病理参数的关系。FISH技术观察cortactin编码基因CTTN在结直肠组织中的扩增。体外合成cortactinsiRNA后转染结直肠癌细胞株SW620,用Westernblot和免疫荧光观察转染后cortactin表达量的变化及细胞分裂状态。结果Cortactin在结直肠癌组织中的表达明显高于腺瘤性息肉和癌旁结直肠正常组织。在结直肠癌组织中cortactin表达增高与结直肠癌不良分化(P〈0.01)、有淋巴结转移(P=0.006)、TNM分期(P=0.022)相关。Cortactin阴性表达或者低表达组结直肠癌患者术后5年生存率明显高于cortactin高表达组。结直肠癌组织中cortactin编码基因CTTN扩增明显,结肠腺瘤与癌旁正常组织中未见扩增。导入cortactinsiRNA的结直肠癌细胞cortactin蛋白表达下降。结论Cortactin异常表达可能与结直肠癌的生长、侵袭及转移有关。Objective To investigate cortactin expression in malignant colorectal tissues and corresponding adjacent non-cancerous colon tissues, precancerous lesions (adenomatous polyps ) and the relationship between the expression of eortactin and cell division in coloreetal cancer cells. Methods The expression of cortaetin was detected by immunohistoehemistry in coloreeta| cancer , coloreeta| adenomatous polyp (precancerous lesions) and coloreetal tissues adjacent to adenoearcinomas (normal tissues). Kaplan- Meier method was employed to compare the survival between the groups. Cortactin expression and cell division were detected by Western blot and immunofluorescence in SW-620 colon cancer cells treated with cortactin siRNA. Results The positive expression rate of eortactin was significantly higher in colorectal cancer tissues than in adenomatous polyp tissues and pericarcinomatous normal tissues. Overexpression of cortactin in colorectal cancer tissues was correlated with poor differentiation ( P 〈 0. 01 ) , lymph node metastasis ( P = 0. 006 ) , and TNM stage ( P = 0. 022 ). The 5 year survival rate of the group of negative/ weak positive expression of cortactin was significantly higher than the group of strong positive expression of eortactin. CTTN gene amplification in coloreetal cancer tissues was obvious. Cortaetin siRNA induction caused a lower cortactin protein expression in colorectal cancer cells. Conclusions It is suggested that the excessive expression of cortactin contributes to the growth of cancer cells in colorectal cancer.
