摘 要:目的:探测生长因子基因在兔关节软骨细胞损伤后不同时间点的表达变化.方法:通过机械划伤制备关节软骨细胞损伤模型,观察软骨细胞在划伤后的形态变化.采用实时荧光定量PCR技术检测软骨细胞在划伤前和划伤后1、3、7d4个时间点的血管内皮生长因子(VEGF)、碱性成纤维生长因子(bFGF)、转化生长因子-β1 (TGF-β1)、胰岛素样生长因子-Ⅰ(IGF-Ⅰ)和Ⅱ型胶原基因mRNA表达.结果:细胞划伤后划痕两侧细胞逐渐向划痕区生长和融合.VEGF和bFGF基因在细胞划伤后呈明显低水平表达,而TGF-β1、IGF-Ⅰ和Ⅱ型胶原基因却呈现较高表达水平.VEGF基因在细胞损伤第3天的表达达高峰后开始下降,而bFGF、TGF-β1及Ⅱ型胶原基因在损伤的第1天表达明显降低,随后呈现升高的趋势.IGF-Ⅰ基因在损伤初期表达变化平稳,在损伤第3天后出现明显升高趋势.结论:bFGF、TGF-β1、VEGF、IGF-Ⅰ和Ⅱ型胶原基因细胞损伤后的表达存在差异性,这为调节关节软骨内源性生长因子基因表达治疗关节软骨损伤提供实验依据.Objective: To explore the growth factors gene expressions in articular chondrocytes of rabbit at different time points after injury. Methods: The scratch-wound model of chondrocyte was established by mechanical injury, and morphological characteristics of chondrocytes were observed. The gene mRNA expressions of vascular endothelial growth factor (VEGF), basic fibroblast growth factor (bFGF), transforming growth factor-β1 (TGF-β1), insulin-like growth factor-I (IGF-I) and type Ⅱ collagen were determined by real time PCR in normal and post-injury 1, 3 and 7 days. Results: The injuried chondrocytes grew gradually and fused from scratch edge to scratch area. The levels of VEGF and bFGF mRNA expression in chondrocytes were decreased after injury, but the expression levels of TGF-β1, IGF-I and type Ⅱ collagen mRNA were increased. The VEGF expression level reached the peak at 7 days after injury and was decreased gradually in the following days. The bFGF, TGF-β1 and type Ⅱcollagen genes mRNA expressions were significantly lowered at day 1 after injury, and then appeared a rising trend. The expression of IGF-I mRNA was smoothly in the early period of cell injury, and increased obviously at 3 days after injury. Conclusion: The expression patterns of bFGF, TGF-β1, VEGF, IGF-I and collagen type Ⅱ mRNA are different after articular chondrocyte injury, which can provide an important basis for molecular biology and the treatment of articular cartilage injury.
