摘 要:目的:探讨山楂叶总黄酮(TFHL)对血管性痴呆大鼠空间学习记忆障碍的改善作用及对脑内胆碱能系统功能活性的影响.方法:健康雄性SD大鼠80只,随机分为Sham、模型组、TFHL 70 mg/kg组、TFHL 140 mg/kg组和盐酸多奈哌齐0.52mg/kg组,每组16只.采用永久性双侧颈总动脉结扎法制备血管性痴呆大鼠模型,TFHL 70、140mg/kg及盐酸多奈哌齐0.52mg/kg组连续给药36 d.采用Morris水迷宫评价大鼠的空间学习记忆能力,H-E染色观察大鼠海马CA1区神经元形态及密度,酶联免疫法检测大鼠皮质、基底节、海马乙酰胆碱酯酶(AchE)和胆碱乙酰基转移酶(ChAT)活性变化.结果:与Sham比较,血管性痴呆模型组大鼠学习记忆能力显著降低,大鼠逃避潜伏期明显延长,穿越平台区次数显著减少,平台象限游泳距离百分比显著降低;ChAT活性明显降低,AChE活性显著增强.TFHL组缩短大鼠逃避潜伏期,增加大鼠穿越平台次数及平台象限游泳距离百分比,增强ChAT活性,降低AChE活性.结论:TFHL可改善血管性痴呆大鼠胆碱能神经系统的功能,从丽提高血管性痴呆大鼠的学习记忆功能.Objective: To investigate the effect of total flavone of hawthorn leaf (TFHL) on the therapeutic effect and mechanism of spatial learning and memory impairment of the vascular dementia rat. Methods: 80 SD male rats were randomly divided into a sham control group, a model group, TFHL70, 140 mg/kg group and donepezil 0.52 mg/kg group (n= 16). Permanent bilateral carotid artery ligation was prepared to establish a vascular dementia model. TFHL70, 140 mg/kg group and donepezil 0.52 mg/kg group were administered for consecutive 36 days. Morris water maze was used to evaluate learning and memory abilities of rats, and colorimetric method was employed to detect cortex, basal ganglia and hippocampus AChE and ChAT content. Results: Compared with the sham control group, learning and memory capacity of the vascular dementia model group significantly reduced, and rat escape incubation period prolonged. The number of crossing the platform area was significantly reduced and the percentage of target quadrant distance was lowered; ChAT activity was significantly lowered, and AChE activity was significantly increased. TFHL rats with vascular dementia improved learning and memory ability, shortened the rats escape latency, increased the number of rat across the platform and the platform quadrant percentage of swimming distance, enhanced ChAT activity, and reduced AChE activity. Conclusion: TFHL can improve vascular dementia rat learning and memory impairment, enhance ChAT activity, reduce the activity of ACHE, increase the acetylcholine content of the brain tissues, and restore cholinergic nerve system function related to chronic cerebral ischemia in rats.
