摘 要:为建立早期快速检测非洲马瘟抗体的IgM捕获ELISA(MAC-ELISA)方法,本试验利用大肠杆菌表达出具有良好抗原性的VP7蛋白,并制备该蛋白的多克隆抗体,通过优化抗原浓度、血清稀释度及酶标抗体稀释度等,建立了检测非洲马瘟的MAC-ELISA方法,并进行了初步应用。结果表明,利用重组表达的VP7蛋白成功建立了检测非洲马瘟的MAC-ELISA方法。The study was aimed to develop an IgM capture enzyme-linked immunosorbent assay for detecting African horse sickness (AHS) . The AHSV VP7 protein with good immunogenicity was expressed in BL21 and polyclonal an- tibody against protein VP7 of African horse sickness virus was prepared. An IgM capture enzyme-linked immunosorbent assay for detecting AHS was developed by optimizing the antigen concentration, the second antibody concentration and the serum dilution. Preliminary application of the MAC-ELISA method indicated that the MAC-ELISA with the recom- bined VP7 protein was successfully developed for detecting antibodies to AHSV with good sensitivity and specificity.
