摘 要:目的:探讨人参皂苷Rg1对染铅大鼠学习记忆保护作用与大脑皮层胶质细胞源性神经营养因子(GDNF)阳性细胞的变化关系。方法:将36只健康3周龄Wistar大鼠随机分为3组:对照组,模型组,Rg1组。采用腹腔注射方式给幼鼠染毒和药物干预。分别采用Y-迷宫实验、原子吸收、免疫组化技术检测各组动物学习记忆能力、血和脑铅含量、大脑皮层GDNF阳性细胞数目。结果:与对照组和Rg1组相比,染铅组大鼠学习记忆能力明显下降(P<0.05),血铅和脑铅含量明显升高(P<0.01)。免疫组化结果显示,染铅大鼠大脑皮层GDNF阳性细胞数目明显增多(P<0.05)。但Rg1组与对照组之间无明显差别。结论:染铅对学习记忆功能影响可能与大脑皮层GDNF阳性细胞的变化有关。Rg1对铅引起大脑皮层GDNF阳性细胞的变化有明显保护作用。Objective : To investigate the relationship between the protection of Ginsenoside Rgl on learning and memo- ry of lead-exposed rats and glial-derived nurotrophic factor (GDNF) ceils. Methods : 36 3 - week-old Wistar rats were randomly divided into 3 groups (n = 12) :control group,lead-exposed group and ginsenoside Rgl administration group. 30 mg/kg lead acetate was used for peritoneal injection to establish the lead poisoning model. Ginsenoside Rgl was perito- neal injected for 14 days. The learning and memory ability was studied with Y-maze test. The blood and brain lead con- centrations were measured with atomic absorption spectrometry. The number of brain GDNF positive cells was observed with immunohistochemistry. Results: Compared with the control and Ginsenoside Rgl groups, the learning and memory a- bility of lead-exposed rats was significantly decreased (P 〈 O. 05 ). The blood and brain lead content in lead-exposed rats was significantly increased (P 〈 0.01 ). The number of GDNF positive cells in brain of lead-exposed rats was obviously increased (P 〈 0.05). But no significant difference was found between the control and Ginsenoside Rgl groups in brain. Conclusion : The effects of lead on learning and memory might be related to the changes of GDNF cells s in brain of lead- exposed rats. Ginsenoside Rgl might play an important role in preventing from lead-induced damages.
