摘  要:目的探讨晚期乳腺癌患者的癌细胞对多西紫杉醇、长春新碱、阿霉素、顺铂等化疗药物的敏感性。方法收集2007年至2012年晚期乳腺癌患者的乳腺癌组织共58例。在无菌条件下剪成两部分,一部分采用胶原酶消化法取得乳腺癌原代细胞并进行体外原代培养,研究多西紫杉醇、长春新碱、阿霉素、顺铂在0.2、0.5、1.0、1.5、2.0PPC 5个浓度下对乳腺癌原代细胞生长的影响,用MTT法检测细胞生长抑制率。另一部分制成石蜡切片,用SP法检测多药耐药基因(MDR1)、耐药相关蛋白(MRP)的表达情况。结果①与MDR1阳性的乳腺癌相比,多西紫杉醇、长春新碱、阿霉素对MDR1阴性的乳腺癌细胞的抑制作用强(P<0.05);而顺铂的抑制作用无明显差异(P>0.05)。②与MRP阴性的乳腺癌相比,四种药物中只有多西紫杉醇对MRP阳性的乳腺癌细胞的抑制作用无明显差异(P>0.05)。结论顺铂对MDR1阳性的乳腺细胞的敏感性较好,多西紫杉醇对MRP阳性的乳腺癌细胞的敏感性较好。Objective: To investigate the sensitivity of advanced breast cancer cells to docetaxel, doxorubicin, vincristine, cispla- tin. Methods: 58 advanced breast cancer tissues were collected from 2007 to 2012. Every tissues was cut into two parts under sterile conditions, one part of specimen were digested by coUagenase and then primary breast cancer cells were cultured in vitro, to study the effects of docetaxel~, doxorubicin, vincristine, cisplatin in 0. 2, 0. 5, 1, 1.5, 2. 0PPC 5 concentration on breast cancer cell growth. cell growth inhibition was tested by MTI" method. Another specimen is made into paraffin sections, The multidrug resistance gene (MDR1), muhidrug resistance associated protein (MRP) were detected by immunohistochemistry SP method. Results: 0)Compared to the breast cancer withMDR1 positive expression , docetaxel, do^ombicin, vincristine have more stronger inhibition to the growth of breast cancer cells with MDR1 negative expression (P 〈 0. 05 ). there was no obviously difference to cisplatin (P 〉 0. 05). (~)Com- pared to the breast cancer with MRP negative expression , only docetaxel have no different inhibition to the growth of breast cancer cells with MDR1 negative expression (P 〉 0. 05). Conclusion : The breast cancer cells with MDR1 positive expression have better sensi- tivity to cisplatin, the breast cancer cells with MRP positive expression have better sensitivity to docetaxel.
