摘 要:以玉米秸秆水解液为原料,对一株产琥珀酸放线杆菌(Actinobacillus succinogenes)ATCC 55618的发酵条件进行优化,研究了培养温度、发酵培养基的初始pH以及培养基成分对该菌株产琥珀酸发酵的影响,并利用响应面法(RSM)对发酵培养基中的水解糖、玉米浆和Na2CO3添加量3个主要因素进行优化,经过优化后的发酵条件为培养温度37℃、培养基初始pH6.8;水解糖质量浓度60g/L、玉米浆质量浓度10g/L、Na2CO3(10mol/L)添加量2mL。在1L的厌氧甁中进行了分批发酵放大实验,琥珀酸产量最高达19.66g/L。Using corn stalk hydrolysate as raw material, the fermentation conditions of Actinobacillus succinogenes ATCC 55618 were optimized. The effect of culture temperature, initial pH, and medium composition on the production of succinic acid was studied, and the addition of hydrolysised sugar, corn syrup and Na2CO3 was optimized by response surface methodology. Results showed that the optimal fermentation was determined as follows: temperature 37℃, initial pH 6.8, hydrolysised sugar mass concentration 60 g/L, corn syrup mass concentration 10 g/L, and 10 mol/L Na2CO3 2 ml. Scale-up experiments were performed in 1 L anaerobic bottle, and the yield of succinic acid could reach 19.66 g/L.
