摘 要:目的:优化好氧反硝化细菌N22’的种子培养基,提高对数期末期细菌浓度。方法:采用Plackett-Burman设计对影响N22’细菌浓度的因素进行评估并筛选出具有显著效应的因素KNO3(X3)、KH2PO4(X4)和K2HPO4(X5),经过最陡爬坡实验接近3个因素的最大响应区域后,应用Box-Behnken设计和响应面分析法确定3个因素的最优水平。结果:优化后的种子培养基:柠檬酸钠6g,KNO32.72g,KH2PO41.35g,K2HPO41.12g,MgSO4·7H2O 0.25g,CaCl20.025g/L,FeSO4·7H2O 0.025g,EDTA0.125g/L;蒸馏水1 000mL,初始pH值7.0。优化后发酵液对数期末期细菌浓度达到1.684 0×1012cfu/mL,比优化前1.632 7×1011cfu/mL提高了9.31倍。结论:Plackett-Burman设计结合响应面分析方法优化了菌株N22’的种子培养基。Objective: The seed culture medium was optimized to improve the bacterial concentration of strain N22'. Method: Firstly, the effects of different factors on bacterial concentration in the seed culture medium was investigated by Plackett - Burman design, Statistically significant factors are : KNO3 ( X3 ), KH2 PO4 ( X4 ) and K2 HPO4 ( X5 ). Then, the steepest ascent experiment was adopted to approach opti- mal region for the three factors. Finally, Box - Behnken design and response surface methodology was used to identify the optimal level of the three factors. Result :The optimal seed culture medium is: sodium citrate 6g, KNO3 2. 72g, KH2PO4 1.35g, K2HPO4 1.12g, MgSO4 · 7H2 0 0.25g, CaCl2 0. 025g, FeSO4 · 7H2O 0. 025g, EDTA 0. 125g;distilled water 1 000mL,initial pH value 7. 0. The bacterial con-centration reached 1.684 0 x 1012 cfu/mL at the telophase of logarithmic phase after optimization, which increased 931% than before. Condusion: The seed culture medium of strain N22' can be optimized through Plackett - Burman design combined with response surface analysis.
