摘  要:目的探讨中成药滋肾育胎丸对培养的滋养细胞凋亡的影响及其机制。方法酶解法分离人足月妊娠胎盘组织中滋养层细胞,培养原代滋养细胞。将体外培养的滋养细胞分为正常对照组、缺氧环境组、低氧环境下不同浓度滋肾育胎丸组。利用流式细胞术(FCM)检测滋养细胞凋亡情况。Western blot法检测体外培养的滋养细胞Fas/FasL蛋白表达量。结果低氧环境组细胞凋亡率较正常组显著增高(P<0.05);低氧环境组与低浓度滋肾育胎丸组细胞凋亡率比较无显著性差异,中、高浓度滋肾育胎丸组细胞凋亡明显低于低氧环境组(P均<0.05)。低氧环境组较正常组FasL蛋白表达增强,较中、高浓度滋肾育胎丸组FasL蛋白表达减少。结论低氧环境可诱导体外培养的滋养细胞凋亡,滋肾育胎丸呈剂量依赖性抑制低氧环境诱导的滋养细胞凋亡,这种作用的机制可能是通过促使滋养细胞下调FasL蛋白表达实现的。'Objective It is to explore the effect of Chinese patent drug Zishen Yutai Pill on apoptosis of cultured human trophoblast cells and its mechanism.Methods Cytotrophoblasts were prepared from term human placenta digested by enzymolysis approach,then the cytotrophoblast were cultured and divided into 3 groups; normal control group,hypoxia conditions group and hypoxia conditions with Zishen Yutai pill plus treatment group.Apoptosis of the cells were detected by flow cytometry (FCM).The expression of Fas/FasL protein in the cells was detected by Western blot method.Results The apoptotic rate of the cytotrophoblast was significantly higher in hypoxia conditions compared with the normal controls (P 〈 0.05).The medium and high concentranon solutions of Zishen Yutai Pill had an inhabit effect on the apoptotic of cytotrophoblast(P 〈 0.05).There was no significant change in low concentration solution of Zishen Yutai Pill (P 〉 0.05).The expression of FasL proteins in hypoxia group was higher than those in the normal controls,lower than that in the medium and high concentration solutions of Zishen Yutai Pill groups.Conclusion Hypoxia induces apoptosis in cultured cytotrophoblasts.The apoptosis of human tropoblasts cells which is induced by hypoxia conditions could be inhibited by Zishen Yutai Pill in dosage-dependent manner,this function my be realized by decreasing the FasL protein expression of the tropoblasts.
