摘  要:目的定量分析非小细胞肺癌组织中TGF-β1和PTEN的表达特点及二者的相关性。方法采用免疫组化SP法检测110例非小细胞肺癌组织和19例正常肺组织中TGF-β1和PTEN的表达,并用图像分析软件定量测试TGF-β1和PTEN蛋白表达的阳性单位(PU)。结果 TGF-β1在非小细胞肺癌组织中的表达高于正常肺组织(P=0.000),TGF-β1的表达与TNM分期及淋巴结转移相关(P值分别为0.000、0.002),而与患者年龄、性别、吸烟史、肺癌大体类型、分化程度和组织学分型未呈现相关性(P>0.05)。PTEN蛋白在非小细胞肺癌组织中的表达明显低于正常肺组织(P=0.000),与非小细胞肺癌的TNM分期有关(P=0.003),而与非小细胞肺癌患者的年龄、性别、吸烟史、肺癌大体类型、分化程度、组织学分型以及淋巴结转移等临床病理特征未呈现相关性(P>0.05)。随着非小细胞肺癌分期的进展,TGF-β1的表达量有不同程度的增强,PTEN的表达量则逐渐减弱,PTEN和TGF-β1的表达呈负相关(r=-0.380)。结论非小细胞肺癌组织中TGF-β1的过表达可能导致PTEN的表达减少,促进非小细胞肺癌的发生发展;TGF-β1和PTEN的联合检测可作为反映非小细胞肺癌生物学行为的指标之一。Objective To investigate the quantitative relationship between the expression of TGF-β1 and PTEN in non-small cell lung cancer tissues. Methods Immunohistochemical SP staining for TGF-β1 PTEN proteins was carried out in 110 cases of non-smallcell lung cancer and 19 cases of normal and lungtissues. The expression intensity (positive unit, PU) of TGF-β1 and PTEN expression was assessed quan- titatively by an image analysis system. Results The intensity of TGF-β1 protein cantly higher in non-small cell lun pression of TGF-β1 was significan g tl were 0. 0000 and 0. 002, respective cancer tissues than that in normal lung tissues Y ly exp (P ression was signifi- = 0. 000). The ex- correlated with TNM stages and lymph node metastases (P values ). There were no association among TGF-131 protein expression and age of patients, gender, history of smoking, degree of differentiation, tumor types and histological types (P 〉 0.05). PTEN positive expression in non-small cell lung cancer tissues was significantly lower than that in normal lung tissue (P = 0. 000), wbich was related to TNM stages (P = 0. 002) and had no signifi- cant relationship with age of patients, gender, history of smoking, degree of differentiation, tumor types, histological types and lymph node metastases (P 〉 0.05 ). Quantitative expression of TGF-JS1 increased with the progression of TNM stages, while expression of PTEN decreased. PTEN was negatively correlated with TGF-β1 expression in non-small cell lung cancer tissues(r = - 0. 380). Conclusions The abnor- mal activation of TGF-β1 protein may lead to a decrease or deletion of PTEN expression in non-small cell lung cancer tissues, which plays an important role in promoting the initiation and development of non- small cell lung cancer. Combined detection of TGF-β1 and PTEN may act as indicators in reflecting the biological behavior of non-small cell lung carcinoma
