摘 要:目的研究大胡蜂肥大细胞脱粒肽12a对血管生成的影响。方法利用鸡胚绒毛尿囊膜(CAM)实验体内观察不同浓度的12a对血管形成的影响;采用MTT法和小管形成实验体外观察不同浓度的12a对人脐静脉内皮细胞(HUVEC)增殖和血管形成的影响。结果 0.1和1μg 12a能明显抑制0.2μg重组人碱性成纤维细胞生长因子(rh-bFGF)诱导的10 d龄鸡胚的绒毛尿囊膜血管生成,导致不同程度的CAM血管大范围明显褪色,密度减少,结构模糊,分支多处断开,生长抑制率分别为60.2%和90.3%;1 mg·L-1和10 mg·L-112a对HUVEC的增殖和血管形成分别有55.4%、39.3%和51.6%、26.7%的抑制率;结论大胡蜂肥大细胞脱粒肽12a能明显抑制新生血管的生成。Aim To investigate the anti'angiogenic effect of 12a, the mastoparan-like peptide from wasp (Vespa magnifica) venom. Methods The tube formarion and proliferation of human umbilical vein endothelial cells (HUVEC) and chicken chorioallantoic membrane (CAM) model were used to observe the anti-angiogenic effect of 12a in vitro and in vivo, respectively. Results In the CAM model, 0.1 ug or 1 ug of 12a could markedly inhibit angiogenesis induced by 0.2 ug rh-bFGF with heavy loss of color, decreasing density and obscure frame of the vessels. The inhibition rates of angiogenesis were 60.2 % for 0.1 ug and 90.3 % for 1 ug, respectively. Accordingly, in HU- VEC culture experiment, the proliferation and angiogenesis of HUVEC treated by 1 mg . L-1 and 10 mg . L-1 of 12a were decreased by 55.4 %, 39.3 % and 51.6 %, 26. 7 %, respectively. Conclusion 12a has a significant anti-angiogenic effect in a concentration-dependent manner.
