摘 要:目的研究缺血后适应对局灶性脑缺血再灌注大鼠外周血淋巴细胞DNA损伤的影响。方法30只雄性SD大鼠随机分为假手术组、脑缺血再灌注组和缺血后适应组。建立大鼠大脑中动脉栓塞模型。假手术组线栓插入后立即拔出;脑缺血再灌注组大鼠脑缺血60min后拔出线栓;缺血后适应组大鼠脑缺血60min后,再灌注20S、再缺血20s,反复5次后恢复再灌注。于术后24h、72h进行神经功能评分。应用单细胞凝胶电泳检测大鼠外周血淋巴细胞DNA的损伤情况。结果与假手术组比较,脑缺血再灌注组和缺血后适应组大鼠术后24h、72h神经功能评分明显降低,外周血淋巴细胞DNA单链与双链损伤均明显增加(均P〈0.01)。与脑缺血再灌注组比较,缺血后适应组大鼠术后24h、72h神经功能评分均明显增高,外周血淋巴细胞DNA单链与双链损伤均明显减少(均P〈0.01)。与术后24h比较,脑缺血再灌注组和缺血后适应组大鼠术后72h外周血淋巴细胞DNA单链与双链损伤均明显减少(P〈0.05~0.01)。结论缺血后适应可减轻大鼠局灶性脑缺血再灌注损伤后外周血淋巴细胞DNA的损伤,具有神经保护的作用。Objective To investigate the effect of ischemic postconditioning on peripheral blood lymphocyte DNA damage after focal cerebral ischemia reperfusion in rats. Methods Thirty male SD rats were randomly divided into sham operation group (SO group), ischemia reperfusion group (IR group) and ischemic postconditioning group (IP group). The middle cerebral artery occlusion models of rats were made. The suture-embolus was pulled out immediately after inserting in SO group; the suture-embolus was pulled out at 60 rain after ischemia in IR group; while in IP group, at 60 min after ischemia, rats were reperfused for 20 s then ischemic for 20 s, repeated five times and at last reperfused. Neurologic scores were evaluated at 24 h and 72 h after operation. The peripheral lymphocytes DNA damage of rats were examined by high performance liquid chromatography. Results Compared with SO group, the neurologic scores of rats at 24 h and 72 h after operation in IR group and IP group were significantly lower, and the DNA single-strand breaks and double-strand breaks of peripheral blood lymphocytes were significant in- creased (all P 〈 0. 01 ). Compared with IR group, the neurologic scores of rats at 24 h and 72 h after operation in IP group were significantly higher, and the DNA single-strand breaks and double-strand breaks of peripheral blood lymphocytes were significantly decreased ( all P 〈0.01 ). Compared with 24 h after operation, the DNA singlestrand breaks and double-strand breaks of peripheral blood lymphocytes in IR group and IP group at 72 h after operation were significantly decreased ( P 〈 0. 05 - 0.01 ). Conclusion Ischemic postconditioning can reduce the peripheral blood lymphocyte DNA damage after focal cerebral ischemia reperfusion in rats, which has protection effects on neurons.
