摘 要:目的运用三种不用方法测定杨树芽提取物体外抗氧化活性及其对DNA损伤的保护作用。方法通过DPPH法、邻苯三酚自氧化法和CuSO4-Phen-Vc-H2O2化学发光体系测定其体外抗氧化活性,并用CuSO4-Phen-Vc-H2O2-DNA化学发光体系测定PBE对·OH致DNA损伤的保护。结果浓度在0.05~0.8mg/mL时,对DPPH自由基的清除率为38.1%~94%;浓度为0.1mg/mL时,随物质的量的增大,对邻苯三酚产生的·O2-抑制率为37.4%~44.2%;浓度在0.05mg/mL^0.8mg/mL时,对·OH自由基的清除率为40.8%~60%。DNA损伤体系中,各浓度均表现出良好的损伤保护作用,浓度大于0.4mg/mL时,效果更加明显。结论杨树芽提取物有较强的抗氧化活性,对DPPH自由基、·O2-和·OH有较强的清除作用,并对·OH所致的DNA损伤有保护作用。Objective Using three different methods for the determination of antioxidant activity and DNA damage preventing effect of poplar bud extracts. Methods By DPPH method, the Pyrogallol autoxidation and CuSO4-Phen-Vc-H2O2 chemiluminescence system for the determination of its antioxidant activity in vitro and chemiluminescent determination of protection of PBE on .OH induced DNA damage by CuSOg-Phen-Vc- H2O2-DNA system. Results At the concentration of 0.05-0.8mg/mL, the DPPH radical scavenging rate of 38.1%-94%; when the concentration of 0. 1 mg/mL, with the increase of the amount of substance of pyrogallol produced · O2inhibition rate was 37.4% - 44.2%; at the concentration of 0.05mg/ml-0.8mg/mL, the ·OH free radical scavenging rate of 40.8%-60%.DNA damage system, each concentration showed a good protective effect, while the concentration is over 0.4mg/mL, the effect is more obvious. Conclusion Poplar bud extracts had strong antioxidative activity, and also had strong scavenging effects on DPPH radical, ·O2 and ·OH, and · OH induced DNA damage had a protective effect.
