非小细胞肺癌组织中EML4-ALK融合基因与TYMS mRNA表达的关系

摘　　要：目的探讨非小细胞肺癌(nonsmall-cell lung cancer,NSCLC)组织中棘皮动物微管样蛋白4-间变淋巴瘤激酶(echinoderm microtubule associated protein like 4-anaplastic Lymphoma kinase,EML4-ALK)融合基因与胸苷酸合成酶(Thymidylate synthase,TYMS)mRNA表达的关系。方法应用实时荧光定量PCR方法检测257例NSCLC组织中EML4-ALK融合基因、TYMS mRNA的表达。结果非小细胞肺癌组织中EML4-ALK融合基因阳性率占4.28%(11/257),在不吸烟患者中较高(P<0.05);TYMS mRNA高表达占63.42%(163/257)。与未检测到EML4-ALK融合基因阳性的非小细胞肺癌患者比较,EML4-ALK融合基因阳性与TYMS mRNA表达水平相关(P<0.05)。结论非小细胞肺癌组织中EML4-ALK融合基因阳性患者TYMS倾向低表达,可能从一线化疗药的培美曲塞中受益。Objective Explore echinoderm microtubule in nonsmall-cell lung cancer tissue-like protein 4-anaplastic Lymphoma kinase echinoderm mierotubule associated protein like 4-anaplastie Lymphoma kinase Fusion gene and Thymidylate synthetase relationship between mRNA expression. Methods Application of real-time fluorescent quantitative PCR method for detection of 257 cases of nonsmall-cell lung cancer echinoderm microtubule associated protein like 4-anaplastic Lymphoma kinase fusion gene, Thymidylate synthase mRNA expression in the tissue. Results echinoderm microtubule associated protein like 4-anaplastic Lymphoma kinase fusion gene-positive rate in nonsmall-eell lung cancer tissue （4.28%）, not in smokers is higher （ P 〈 0. 05 ） ; Thymidylate synthase mRNA overexpression （ 63. 42% ）. With the echinoderm microtubule associated protein like 4-anaplastic Lymphoma kinase fusion gene was not detected-positive nonsmall-cell lung cancer compared with echinoderm microtubule associated protein like 4-anaplastie Lymphoma kinase fusion gene-positive and Thymidylate synthase mRNA expression levels （ P 〈 0.05 ）. Conclusions In nonsmall-cell lung cancer tissue echinoderm microtubule associated protein like 4-anaplastic Lymphoma kinase Thymidylate synthase low expression of fusion gene-positive patients may benefit from first-line chemotherapy drugs in Pemetrexeddisodium.
